摘 要:目的:观察微弧氧化(MAO)方法对近β钛合金Ti-5Zr-3Sn-5Mo-15Nb(TLM)表面处理后对成骨细胞早期行为的影响.方法:用脉冲直流电源在两个不同电压下对TLM表面进行微弧氧化处理,并用电镜观察试样表面形貌.MTT方法检测成骨细胞增殖,用商业试剂盒检测总蛋白合成及碱性磷酸酶(ALP)活性.结果:微弧氧化处理可以在TLM表面形成一层多孔生物活性氧化层,其形貌与微弧氧化电压有关,低电压时形成的孔洞结构较小,高电压时形成的孔洞结构较大.成骨细胞在微弧氧化表面的早期增殖(24,72,120h)及7d细胞总蛋白合成明显高于抛光表面,且细胞在高电压微弧氧化表面的增殖优于低电压表面.而培养7d后微弧氧化表面的细胞ALP活性明显低于抛光表面,且高电压表面的最低.结论:微弧氧化处理改变了TLM表面形貌以及其它特性,从而促进成骨细胞在其表面的增殖及合成分泌活性,但抑制其ALP活性.不同电压下形成的不同微弧氧化形貌对成骨细胞的早期行为有影响.AIM: To modify the surface of a novel near β titanium alloy Ti-SZr-3Sn-SMo-15Nb ( TLM ) with micro-arc oxidation ( MAO), and to evaluate the initial responses of osteoblasts to the modified surfaces. METHODS: MAO of the specimens was carried out using a pulsed DC power supply at two different voltages, and the morphology of specimens was observed using scanning electron microscope. MTT method was used to evaluate cell proliferation, meanwhile total intracellular protein synthesis and alkaline phosphatase (ALP) activity were assessed by commercial available kits. RESULTS: A porous bioactive oxide layer was grown on TLM substrates via MAO process, and the diameters of pores were positively correlated to the voltages applied. Initial osteoblast proliferation (24, 72 and 120 h) and total intracellular protein synthesis (7 d) were observed more significantly on the micro-arc oxidized surfaces than on the unmodified surface, and osteoblasts proliferation was the best on the micro-arc oxidized surfaces formed at higher voltage. However, after 7-d culture, ALP activity was lower on the micro-arc oxidized surfaces, and showed the lowest on the micro-arc oxidized surface formed at higher voltage. CONCLUSION: MAO process can alter the surface characteristics of TLM, thus the proliferation and synthesis and secretion activity of osteoblasts on TLM are enhanced, while their differentiation is inhibited. Besides, the different morphologies related to MAO voltages applied exert different effects on initial cell behaviours.
