摘 要:【目的】观察TGF-β1和P-gp、GST-π在大肠癌组织和癌旁组织的表达情况,探讨三者表达与大肠癌临床病理特征和复发转移之间的关系,为临床化疗药物的选择和评估病人的预后提供参考依据。【方法】用免疫组化S-P法检测48例大肠癌组织和45例癌旁黏膜组织中TGF-β1和P-gp、GST-π的表达水平,并分析三者与大肠癌临床病理特征和复发转移之间的关系。【结果】TGF-β1和P-gp、GST-π在大肠癌组织中的表达显著高于癌旁组织;P-gp、GST-π在不同年龄、性别、肿块大小、分化程度、Dukes分期的癌组织中表达均无统计学差异;TGF-β1的表达与肿块大小、Dukes分期有关。复发转移组TGF-β1和P-gp、GST-π的表达高于未复发转移组,两者相比有统计学差异;P-gp与GST-π,P-gp与TGF-β1表达存在相互关联性。【结论】检测大肠癌中TGF-β1和P-gp、GST-π三种指标似可作为大肠癌预后和化疗方案设计的指标。[Objective]To observe the expressions of transforming growth factor beta I gene(TGF-π ), Pglycoprotein (P-gp) and glutathione S-transferase-π(GST-π) in pericancerous colorectal mucosa tissues and colorectal cancer tissue and to investigate the correlation of the expressions of P-gp, GST-π and TGF-β1 with clinical pathological parameters and recurrence or metastasis in colorectaI cancer. [Methods] The expressions of TGF-β1 , P-gp and GST-π were detected in forty eight cases of colorectal cancer tissue and forty five cases of pericancerous colorectal mucosa tissues by immunohistochemistry. The correlations of P-gp, GST-π and TGF-β1 expression with clinical pathological parameters, recurrence or metastasis in colorectal carcinoma were analyzed, and the correlation of P-gp, GST-π and TGF-β1 expression were analyzed as well. [Results] The expression levels of TGF-β1 , P-gp and GST-π in colorectal cancer tissue were significantly higher than those of pericancerous mucosa tissues. The expressions of P-gp and GST-π in colorectal carcinoma were not associated with patients' age, sex, tumor size, histological grade, infiltration depth, Dukes staging and lymph node metastasis. The expression of TGF-β1 in colorectal carcinoma was not associated with patients" age, sex, histological grade, but associated with tumor size, Dukes staging. The expressions of P-gp, GST-π and TGF-β1 in colorectal cancer tissue with recurrence or metastasis were significantly higher than those without recurrence or metastasis. The expression of P-gp in colorectal cancer tissue was well correlated with that of GST-π, but it was well correlated with that of TGF-β1. There was no correlation between the expression of GST-π and TGF- β1. [Conclusion] Detecting the expressions of TGF-β1 and P-gp , GST-π in colorectal carcinoma may be helpful to predict prognosis and choice of chemotherapy.
