顺铂联合全反式维甲酸对OCM-1裸鼠移植瘤模型作用的研究

摘  要:目的观察顺铂(CDDP)和全反式维甲酸(ATRA)对脉络膜恶性黑色素瘤细胞株OCM-1裸鼠移植瘤的作用。方法采用体外细胞培养法培养OCM-1,20只4周龄雄性裸鼠随机分为4组,将细胞接种至裸鼠皮下,建立异位移植瘤模型。CDDP、ATRA及两药联合分别处理移植瘤模型,TUNEL法检测肿瘤组织。结果CDDP组抑瘤率为43.20%;ATRA组抑瘤率为37.65%;联合组抑瘤率为65.94%,经金氏公式计算得出q值为1.02(1.153>q>0.85)。结论CDDP和ATRA对OCM-1裸鼠移植瘤的抑制具有相加作用。CDDP和ATRA联合应用可抑制肿瘤生长,其机制可能和诱导肿瘤细胞凋亡有关。Objective Uveal melanoma is the most common eye malignant tumor in adult,which has a high mortality rate and responese poorly to the existing chemotherapy. How to reverse drug resistance and improve the prognosis of patients has become an important research point. Therefore the experiment was to study the influence of cis-diamminedichloroplatinum (CDDP) and/or all-trans retinoic acid(ATRA) on growth of xenografts of choroidal malignant melanoma lines( OCM ) in nude mouse. Methods 5 ~ 107/mL of cell suspension from OMC-1 was subdermally injected in 20 male Balb/c nude mice to creat human OCM-1 xenograft model. The model mice were randomly divided into 4 groups. 2 mg/kg of CDDP,20mg/kg of ATRA and combination of 2 mg/kg CDDP and 13.44 mg/kg ATRA was intraperitoneally administrated after 2 weeks of modeling for 4 weeks in group A, B, C respectively, and group D was as the blank control group. The size of tumor were measured before and after usage of CDDP or/and ATRA, and the histological alteration of tumor and apoptosis was examined under the light microscope and TUNEL technology. Results The inhibition rate of CDDP to tumor was 43.2% , and that of ATRA was 37.65% , and the inhibition rate of the group CDDP + ATRA was 65.94%.The q value is 1.02 (1. 153 〉 q 〉 0.85) calculated by Jin' s formula. The mass doubling time of group A was obviously longer than that of blank control ( P 〈 0.05 ). The mass doubling time of group B was longer than that of blank control but without significant difference( P 〉0.05 ). The mass doubling time of group C was obviously longer than that of blank control( P 〈0. 01 ). Through the TUNEL testing,all the tumor tissues of group A ,B and C showed different degree of positive staining. Conclusion The combination of CDDP and ATRA has the stronger inhibition on the growth of xenografts of OCM-1 in nude mice, and its main mechanism may be related to the induction of apoptosis of tumor cells.
