摘  要:目的探讨c-kit信号传导在全反式维甲酸(ATRA)诱导急性早幼粒细胞白血病(APL)细胞株NB4细胞分化中的作用。方法用ATRA诱导APL细胞株NB4。RT-PCR、Western blot分别检测诱导后NB4细胞中c-kit mRNA、c-kit蛋白的表达;流式细胞仪(FCM)测定细胞表面CD117表达情况。结果ATRA诱导后的NB4细胞稳定表达c-kit基因;随细胞的分化成熟,c-kit mRNA及c-kit蛋白的表达呈进行性下降,ATRA处理后24、48、72 h,c-kit mRNA表达分别下降38.9%、44.4%和61.1%,c-kit蛋白分别下降4.3%,34.6%,80.2%;细胞表面CD117表达率极低,ATRA处理前后无明显变化。结论c-kit细胞内信号传导可能参与ATRA诱导NB4细胞分化的调节。Objective To investigate the role of c-kit signaling during cell differentiation of NB4 cells treated with all trans retinoid acid (ATRA). Methods Trypan blue exclusion assay was used for cell count, cell morphology was observed by microscopy,cell differentiation was detected by nitro blue tetrazolium (NBT) reduction test,c-kit mRNA expression was assayed by RT-PCR, expression of c-kit protein was analyzed by Western blot;CD117 expression was detected by flow cytometry. Results Inhibition of differentiation and proliferation were found in NB4 cells by ATRA treatment; expressions of c-kit mRNA and protein were down-regulated during the differentiation of NB4 cells; c-kit mRNA expression showed 38.9%, 44.4%, 61.1% reduction and SCF receptor expression decreased in 4.3%, 34.6%, 80.2% respectively after 24 h,48h and 72 hours in NB4 cells-treated by ATRA. Expression of CD117 in NB4 cells was negative. Conclusions ATRA induces differentiation and maturation of NB4 cells, c-kit mRNA and intracellular protein expressions are down-regulated during the differentiation of NB4 cells, these results suggest that c-kit intracellular signaling may be involved in regulation of NB4 cell differentiation.
