摘  要:目的探讨枸杞多糖(Lycium barbarum polysaccharide,LBP)对人前列腺癌PC-3细胞的影响及其机制,观察LBP对人前列腺癌PC-3细胞致瘤作用的干预效果。方法通过细胞生长抑制率、彗星实验、流式细胞术、原位末端标记法(TUNEL)和Bcl-2、Bax蛋白表达来检测LBP对人前列腺癌PC-3细胞的影响;致瘤性实验检测LBP对人前列腺癌PC-3细胞致瘤力的影响。结果LBP能抑制人前列腺癌PC-3细胞的生长;可致PC-3细胞DNA链断裂;能诱导PC-3细胞凋亡;使Bcl-2/Bax蛋白比值显著下降;LBP干预组肿瘤体积增长缓慢,肿瘤体积和重量均明显小于和轻于人前列腺癌裸鼠移植模型组。结论LBP对抗人前列腺癌PC-3细胞和肿瘤生长抑制作用的机制,可能与其损伤人前列腺癌PC-3细胞DNA链、诱导细胞凋亡和调控凋亡相关基因表达有关。Objective To investigate the influence of Lycium barbarum polysaccharides (LBP) on the growth of human prostate carcinoma PC-3 cells and its mechanism and study the intervention of LBP on induced tumor. Method Trypan blue exclusion staining was used to examine the inhibitory effect of LBP on PC-3 cells. The influence of LBP on the cell cycle and the apoptosis were observed with flow cytometry and TUNEL Single cell gel electrophoresis assay was applied to determine the DNA damage of PC -3 cell. The immunohistochemical technique was used to detect Bcl-2 and Bax expression. The induced tumor test was used to observe the inhibitory effect of LBP on tumor. Results LBP could inhibit the growth of PC-3 cells and induce cells apoptosis. LBP could induce the breakage of DNA strand of PC-3 cells. The ratio of Bcl-2/Bax protein decreased. The growth of tumor was inhibited by LBP. Conclusion The anti-prostate cancer mechanism of LBP may be related to its influence on inducing apoptosis, damaging DNA chain, and adjusting Bcl-2/Bax expression.
