摘 要:背景与目的:已知新发现的半胱氨酸蛋白酶legumain可能参与肿瘤的发生发展。本研究检测了le-gumain在荷瘤组织及荷瘤与正常小鼠免疫细胞如脾脏T、B和巨噬细胞中的差异表达,为研究legumain在抗肿瘤免疫中的作用提供基础数据。方法:取正常和4T1荷瘤的雌性BALB/c小鼠的肿瘤组织或脾细胞,分离巨噬细胞和T、B淋巴细胞,检测legumain的基因表达水平。结果:在荷瘤小鼠的肿瘤组织和淋巴器官均检测到高表达的legu-main,而且随着肿瘤进展,其表达水平相应提高;legumain除高表达于巨噬细胞外,脾脏中的淋巴细胞,特别是T淋巴细胞,与正常小鼠相比其legumain的表达水平显著升高。结论:荷瘤小鼠的肿瘤组织及其脾脏巨噬细胞和T细胞中高表达legumain,并且与肿瘤的发生发展呈正相关。Background and purpose: Legumain has been reported to be involved in the tumor progression. We investigated the differential expression of legumain in splenic T, B and macropbages from tumor-beating mice to study the possible role of legumain in anti-tumor immunity. Methods: The splenocytes were isolated from normal and 4TI tumor-bearing female BALB/c mice, splenic macropbages were isolated by adhering method, T and B lympbocytes were enriched by the nylon wool. Legumain expression in macropbages, T and B lympbocytes was determined. Results: High expression level of legumain was detected both in the tumor tissue and splenocytes including macropbages, T and B lympbocytes which derived from 4TI tumor-beating mice and the expression level increased as the tumor progressed. Especially, legumain level was significantly higher in T cells from tumor-bearing mice compared to normal mice. Conclusions: Increased expression of legumain during the tumor progression indicated that legumain might be involved in the tumor growth.
