摘  要:目的探讨转化生长因子β(transforming growth factor-β,TGF-β)受体在不同年龄SD大鼠晶状体上皮细胞中的表达变化及意义。方法按1、3、6个月鼠龄分为3组,采用晶状体前囊膜铺片方式,应用荧光免疫组化技术进行TGF-β受体TGF-βR1和TGF-βR2的荧光免疫组化染色,检测其蛋白水平的表达;采用剥取晶状体囊膜组织方法,应用RT-PCR技术检测TGF-βR1和TGF-βR2在转录水平的表达。结果在蛋白水平,TGF-βR1和TGF-βR2于1个月鼠龄组表达最强,IOD分别为478001±44417和778338±62596;3个月鼠龄组表达减弱,分别为360343±33196和360343±33196;6个月鼠龄组表达最弱,分别为225858±23483和274648±29801,组间差异有显著性(P<0.05)。在转录水平,TGF-βR1和TGF-βR2mRNA表达量在1个月龄组分别为3.0±0.9和3.4±1.1;3个月龄组分别为2.1±0.6和2.6±0.8;6个月龄组分别为1.3±0.3和1.6±0.5,随着年龄的增加而减弱(P<0.05)。结论大鼠晶状体上皮细胞中TGF-βR1和TGF-βR2的表达随年龄的增长而减弱,提示对TGF-β的敏感性与年龄呈负相关。Objective To investigate the age-related expression of TGF-β receptors in rat lens epithelial cells. Methods Sprague Dawley rats were divided into three groups based on age:1, 3 and 6 months. An technique was used to detect the expression of TGF-βR1 and TGF-βR2 in rat lens epithelial cells. RT-PCR was applied to identify the expression of TGF-βR1 and TGF-βR2 mRNA in rat lens epithelial ceils. Expression of the TGF-β receptor in the three groups was analyzed. Results The expression of TGF-βR1 and TGF-βR2 at protein and mRNA levels was highest in the 1-month-old group. The expression of TGF-βR1 and TGF-βR2. at protein and mRNA levels was lower in the 3-month-old group than in the 1-month-old group. The expression of TGF-βR1 and TGF-βR2 at protein and mRNA levels in the 6-month-old group was the lowest of the three groups. Condusion In rat lens epithelial cells, there is a significant age-related decline in TGF-βR1 and TGF-βR2 expression at protein and transcription levels.
