摘 要:目的应用链脲佐菌素(Streptozocin,STZ)诱导建立糖尿病大鼠动物模型,观察膳食中添加牛磺酸对糖尿病大鼠视网膜损伤的影响。方法SD大鼠用链脲佐菌素诱导糖尿病,分为正常对照组、糖尿病组、1%牛磺酸干预糖尿病组、5%牛磺酸干预糖尿病组、胰岛素治疗糖尿病组。2个月后测定视网膜电图(electroretinograph,ERG),并应用免疫组织荧光双标化学技术检测视网膜中胶质原纤维酸性蛋白(glial fibrillary acid protein,GFAP)、牛磺酸转运蛋白(taurine trans-porter,Taut)表达。结果视网膜电图结果显示:糖尿病组视杆细胞反应a波、b波、最大反应b波、phot-ERGb波、OPsp4波的潜伏期分别比正常对照组明显延长;OPsOs1波的幅值比正常对照组明显降低;5%牛磺酸干预可明显缩短以上各波的潜伏期,升高OPsOs1波的幅值。免疫荧光结果显示:与正常对照组相比,糖尿病组整个视网膜GFAP表达明显增加,其阳性表达主要出现在外核层、外网状层、内核层、内网状层,视网膜中Taut的表达明显减少,只在外界膜、外核层和外网状层有少量表达;1%牛磺酸和5%牛磺酸干预可降低糖尿病大鼠视网膜中GFAP的表达,增加视网膜中Taut的表达,呈剂量效应关系。结论膳食喂饲牛磺酸对糖尿病引起的视网膜损伤有一定的保护作用,其保护作用可能与降低视网膜中GFAP表达、增加Taut表达有关。Objective To investigate the effects of taurine against the retinal damage in diabetic rats. Methods Sixty male SD rats were randomly divided into 5 groups with 12 rats in each group: normal control group ( NC), diabetes mellitus group ( DM), 1% taurine intervention plus diabetes mellitus group ( DM + 1% T), 5% taurine intervention plus diabetes mellitus group (DM + 5% T), insulin treatment plus diabetes mellitus group ( DM + I). Streptozotocin was used to induce diabetes in rats. Simultaneously, taurine was added into forage at 1% or 5% as taurine intervention and insulin (20 U/kg) was subcutaneously injected as insulin treatment. Two months after induction of DM, the retinal function was detected by electroretinograph (ERG) ; the glial fibrillary acid protein (GFAP) and taurine transporter expression in the rat retina were determined by immunohistofluorescence. Results The implicit time of a and b wave of rod-response, b wave of max-response, p4 wave of OPs and b wave of phot-ERG of diabetic rats was respectively prolonged significantly as compared to that of control rats. The Osl amplitude of OPs was significantly decreased in diabetic rats. The GFAP expression in inner nuclear layer, outer nuclear layer, inner plexiform layer and outer plexiform layer increased 2 months after induction of DM. The taurine transporter expression in retina decreased 2 months after induction of DM. Taurine up-regulated GFAP expression and down-regulated taurine transporter expression in retinal cells.
