摘  要:[目的]通过对照实验探讨自发性癫痫大鼠(spontaneously epilepsy rats,SER)与正常Wistar大鼠海马区胶质细胞型谷氨酸转运体GLAST蛋白的表达情况,以期发现其与遗传性癫痫发病的关系。[方法]利用聚合酶链反应(PCR)扩增SER的特异性基因片段,筛选出SER;利用免疫荧光定位GLAST蛋白的表达,并应用免疫组织化学技术,检测GLAST蛋白在SER及正常Wistar大鼠海马内的表达情况。[结果]GLAST蛋白在SER海马的齿状回(DG)表达与正常Wistar大鼠相比明显减少(GLAST的表达为Wistar:52.67±11.5,SER:39.39±9.48,P<0.05),在海马CA1和CA3区表达差异无显著性意义(CA1区为Wistar:48.56±10.6,SER:45.84±7.20;CA3区为Wistar:43.29±10.6,SER:36.73±6.50,P>0.05)。[结论]SER自发性癫痫可能与海马DG区GLAST蛋白表达下降有关;GLAST表达下降可能是由SER基因位点的突变引起的。[ Objective ] To investigate the expression of glutamate transporters GLAST protein in hippocampus of spontaneously epileptic rats (SER) and further explore the role of GLAST in the occurrence of epilepsy. [Methods] The SER was resieued by DNA extraction and PCR. The expression location of GLAST protein were detected by immunohischemistry and immunofluorensence respectively. [ Results] The GLAST protein expmessien was lower in dentate gyrus (DG) of hippocampus in SER than that in cantral rats. There was not significant difference about the GLAST protein in CA1 and CA3 between SER and cantral rats. [ Conclusion] Down - regulation of GLAST protein is correlated with the occurrence of epilepsy in SER.
