卵巢腺癌组织中化学趋化因子mRNA表达与TAM、MC计数的相关性研究

摘  要:目的研究卵巢腺癌组织中IL-8mRNA、MCP-1 mRNA和MIP-1α mRNA表达水平及肿瘤相关巨噬细胞(TAM)、肥大细胞(MC)计数,探讨它们之间的相互关系及临床病理意义。方法43例卵巢腺癌常规制作石蜡包埋切片,3种化学趋化因子mRNA为原位杂交染色,TAM和MC染色为EnVisionTM免疫组化法。结果组织学分级G1、临床分期Ⅰ+Ⅱ、无淋巴结转移和未侵犯周围组织器官的病例IL-8 mRNA、MCP-1mRNA和MIP-1αmRNA表达阳性率及TAM、MC计数均值明显低于组织学分级G3、临床分期Ⅲ+Ⅳ、淋巴结转移和侵犯周围组织器官病例(P<0.05或P<0.01);IL-8mRNA、MCP-1mRNA和MIP-1αmRNA表达阳性病例TAM、MC计数均明显高于阴性病例(P<0.01)。结论3种化学趋化因子mRNA及TAM、MC计数均为反映卵巢腺癌进展、生物学行为和预后的重要标志物,3种化学趋化因子mRNA均能促进癌基质中TAM、MC浸润和迁移。Objective To study on the expressions of IL - 8mRNA, MCP - lmRNA and MIP - lmRNA and the Counts of TAM and MC and their correlation in ovarian adenocarcinoma tissues. Methods detecting the expressive levels of IL - 8mRNA, MCP - 1mRNA In situ hybridization were derected,and MIP - 1αmRNA and Envision immunohistochemistry wes used for the counts of TAM and MC. Resuits The positive rates of IL - 8mRNA, MCP -1mRNA and MIP-1α mRNA expressions and the counts of TAM and MC were significantly lower in the cases of histologic grade G1, clinical stage Ⅰ+Ⅱ, no - metastasis of regional lymph node, and no - infiltration of peritumoral tissues than those in the ones of histologic grade G3, clinical stage Ⅲ-Ⅳ, metastasis of regional lymph node, and infiltration peritumoral tissues (P 〈 0. 05 or P 〈 0. 01 ). The counts of TAM ,MC were significantly higher in the positive cases of IL- 8mRNA, MCP - 1 mRNA and MIP -1αmRNA than those in the negative cases ( P 〈 0.01 ). Conclusions The expressions of chemakine' s mRNA and the eounts of TAM and MC might be important biological markers for reflecting the progression, biological behaviors and prognosis of ovarian adenoearcinoma. Three chemokines could enhance the infiltration and migration of TAM and MC in neoplastic matrix.
