摘  要:目的研究BH3-only蛋白Bim在紫杉醇诱导胃腺癌细胞凋亡中的作用。方法流式细胞仪PI单染法检测细胞凋亡率;JC-1染色检测线粒体膜电位的变化;在紫杉醇相对敏感SGC-7901胃腺癌细胞株中采用小干扰RNA(siRNA)技术特异性"沉默"Bim基因;Western blot分别检测紫杉醇诱导siRNA"沉默"Bim基因前后Bim蛋白的表达水平变化。结果紫杉醇诱导胃腺癌细胞凋亡具有剂量和时间依赖性,SGC-7901较BGC-823胃腺癌细胞株对紫杉醇更为敏感。紫杉醇诱导SGC-7901及BGC-823胃腺癌细胞很大程度依赖线粒体途径凋亡。特异性"沉默"Bim基因的转录,紫杉醇诱导凋亡敏感细胞株SGC-7901的凋亡率明显降低。结论Bim在紫杉醇诱导胃腺癌细胞凋亡中发挥关键调节作用。Objective To study the role gastric adenocarcinoma cells. Methods of BH3-only proapoptosis protein Apoptosis was measured by flow Bim in apoptosis of paclitaxel-induced cytometry using propidium iodide staining. Changes in mitochondrial membrane potential were studied using JC-1 staining. Bim-specific siRNA and con- trol siRNA was transfected into the sensitive SGC-7901 cells, respectively. Western blot was used to detect the target gene expression at protein levels. Results Paclitaxel induced apoptosis of gastric adenocarcinoma cells in a dose- and time-dependent manner. It appeared that SGC-7901 cells were more sensitive to paclitaxel-induced apoptosis than BGC-823 cells. Paclitaxel induced mitochondrial-dependent apoptosis in SGC-7901 and BGC-823. Inhib- iting Bim expression by siRNA protects gastric adenocarcinoma cells from paclitaxel-induced apoptosis. Conclusion BH3-only protein Bim plays a critical role in paclitaxel-induced apoptosis of gastric adenocarcinoma cells.
