摘 要:建立了定性分析亚麻籽饼粕中开环异落叶松树脂酚二葡萄糖苷(SDG)的TLC方法,根据TLC结果,确定提取溶剂为0.1 mol KOH溶于1 L 70%乙醇,固液比为1∶8,提取2次。然后用大孔吸附树脂与TLC相结合的方法,探讨了亚麻籽中SDG的分离纯化,并根据HPLC分析结果确定了最佳的分离纯化工艺条件。即当亚麻籽饼粕KOH-乙醇-水提取液经中和浓缩后,用DM-21型大孔吸附树脂吸附,水洗去杂质,60%乙醇洗脱,浓缩干燥,经HPLC检测,亚麻籽饼粕含SDG为1.64%,粗提物产率达4.98%,SDG质量分数为32.93%。结果表明:KOH-乙醇-水提取与大孔吸附树脂相结合制备SDG提取物的方法与传统分离法相比,得率高、工艺操作简单且安全无毒。The lignan secoisolariciresinol diglucoside (SDG) is a bioactive molecule effective on anticancer, antiviras and antioxidation. A simple and direct method for SDG qualitative analysis was established with TLC. Suitable macroporous adsorption resin (MAR DM-21 ) was chosen to separate SDG from extract solvent system (KOH-ethanol-water) of flaxseed cake. The identification of SDG and the determination of the purity of targeted fraction were carried out with HPLC. It was found that DM-21 showed an extract rate as 4.98 % and the purified fraction contained 32.93 % (W/W) SDG. Compared to the conventional extraction method,the yield of SDG was improved significantly and the extract time was greatly reduced. It was considered as a safe and suitable medium for sealed-up purification of SDG.
