摘  要:目的研究阻断细胞外信号调节激酶(ERK)通路的再激活对吗啡依赖大鼠已建立的条件性位置偏爱(CPP)的影响。方法将80只大鼠随机分成8组每组各10只,Ⅱ-Ⅷ组为实验组,Ⅰ组为对照组。在建立吗啡的CPP后,分别给予腹腔注射细胞外信号调节激酶抑制剂SL327或生理盐水后再次注射吗啡,对照组仅注射生理盐水,并与给药环境建立联系,观察对已建立的CPP的影响。并采用western blotting法测定伏隔核中磷酸化ERK1/2(P-ERK1/2)的表达水平。结果①CPP测试前各组在白箱内(伴药侧)停留的时间差异无统计学意义(P>0.05)。②经过6d条件反射训练,各实验组大鼠在白箱内停留的时间明显比对照组长,差异均有统计学意义(P<0.01),表明实验组大鼠均已建立起对白盒的CPP。③给予已建立CPP的各实验组大鼠不同处置,结果表明,只有给予腹腔注射SL327后再次注射吗啡的Ⅳ组大鼠的CPP已消除,该组在白箱内的停留时间较其它实验组有显著性差异(P<0.01),而与对照组的差异无统计学意义(P>0.05)。同时,Ⅳ组大鼠伏隔核中P-ERK1/2的表达水平显著低于其它实验组(P<0.01),但其与对照组间的差异无统计学意义(P>0.05)。结论阻断ERK通路的再激活可以消除吗啡依赖大鼠已建立的条件性位置偏爱。Objective To study the influence of blockade of ERK pathway reactivation on the previously learned place preference in the morphine-addicted rats. Methods Eighty rats were randomly assigned into eight groups with ten in each group: the first group was the control group, and the other seven groups were the experimental groups. After the estab- lishment of morphine-producing conditioned place preference ( CPP), the rats in the experimental groups were administered peritoneal injection of morphine after normal saline or SL327 an inhibitor of extracellular signal-regulated kinase ( ERK), while the control animals were only administered normal saline. The relation between injection and surroundings was established and the influence to established CPP was observed. The expression of the phosphorylation of ERK (P-ERK) in nucleus accumbens septum (NAcs) of rats was measured by Western blotting. Results Before the acquisition of the CPP, the difference of the time for rats staying in the white box ( drug side) among all groups was not significant ( P 〉 0. 05 ). The rats in each of the experimental groups significantly stayed longer in the white box than the rats in the control group after the 6-day conditioned behavior training (P 〉 0.05 ), which indicated an established CPP to the white box in all the experimen- tal groups. After the different treatments, the CPP of rats in IV experimental group, which were administered the peritoneal injection of morphine after SL327 and then were placed in the white box, was eliminated. The rats in this group stayed significantly longer in the white box than the rats in the other experimental groups ( P 〈 0. 01 ). However compared with the control group, the difference was not significant ( P 〉 0. 05 ). The expression levels of P-ERKI/2 in rats' NAcs of IV experimental group were significantly lower than those in the other experimental groups (P 〈0. 01 ) but no significantly different from the control group (P 〉 0. 05).
