摘 要:目的研究大鼠实验性大脑内出血后脑组织基质金属蛋白酶-9(matrix metalloproteinases-9,MMP-9)及核转录因子-κB(NF-κB)的表达及变化规律。方法采用立体定向技术将自体不凝血注入大鼠尾状核区制备不同时间段的大脑内出血模型,用免疫组化染色法检测不同出血时间周围脑组织MMP-9及NF-κB的表达。结果MMP-9的阳性细胞表达数在出血后3h出现第一个表达高峰,之后递减,出血后1d再次升高并于出血后2d达到最高峰,之后又下降。NF-κB在出血后12h上升,2d达高峰,之后下降。MMP-9与NF-κB呈低度相关。结论MMP-9在大脑内出血后显著表达并与脑水肿的发生发展有关,NF-κB对MMP-9的表达进行调节。Objective To investigate the expressions of matrix metalloproteinases-9 (MMP-9) and nuclear factor-κB (NF-κB) in rat brain tissue at different time after experimental intracerebral hemorrhage. Methods Rat experimental intracerebral hemorrhage(ICH) model was developed by utilizing stereotactic apparatus and infus- ing antologous blood into the left caudate nucleus. The expressions of MMP-9 and NF-κB in rat brain tissues were detected by immunohistochemistry. Results The first peak of MMP-9 was observed at 3h after ICH, then its expression was decreased. During 12h- 1d after ICH, the expression of MMP-9 was upgraded, and peaked at 2d, then decreased again. The expression of NF-κB was upgraded at 12h and also peaked at 2d after ICH, then it was decreased. The expression of MMP-9 and NF-κB had low correlation. Conclusion The expression of MMP-9 diverses at different time in the brain after ICH according to the dividation of edema development phase in brain, and is regulated by NF-κB.
