摘  要:目的研究树舌多糖抑制肿瘤的作用机制。方法通过随机分组实验,测定树舌多糖对小鼠HepA瘤的抑瘤率,经Western blot检测caspase-8与TNF-α的表达,免疫荧光法检测MAP-2的表达,并通过电镜观察树舌多糖作用后细胞结构的变化。结果树舌多糖的抑瘤率为51.82%,经其作用后,Western blot显示caspase-8、TNF-α表达均显著增加,免疫荧光法检测MAP-2的表达也显著增加。电镜下观察到树舌多糖作用后细胞结构发生改变并形成凋亡细胞。结论树舌多糖通过促进caspase-8的表达,刺激TNF-α表达增加,进一步影响细胞骨架的重排从而促进细胞凋亡抑制小鼠HepA瘤的生长。Objective To investigate the mechanism of Ganoderm applanatum polysaccharide (GAP) against tumor cells. Methods The tumor weight were measured in HepA mice, the expressions of caspase-8 and TNF- α were measured by Western blot; the expression of MAP-2 was measured by immunofluorescenee(IF) ;the tumor cell morphology were observed under electron microscopy. Results The tumor inhibition rote conducted by GAP was 51.82% .After treatment with GAP for 7 days, Western blot showed that the expressions of caspase- 8 and TNF-α increased. Immunofluorescence showed that the expression of MAP-2 increased in the same condition. The changes of the cell structure had been observed in the tumor tissues under EM after GAP treatment. Conclusion GAP can affect reassembly of cytoskeleton, accelerate cell apoptosis and suppress the growth of the HepA mouse tumor by accelerating the expresssions of caspase-8 and TNF-α.
