摘  要:目的通过检测Bcl-2、FasL在实验性自身免疫性葡萄膜炎(EAU)的表达,探讨Fas/FasL介导的细胞凋亡在实验性自身免疫性葡萄膜炎(EAU)发病机制中的作用。方法自新鲜牛眼球中提取S抗原,与弗氏完全佐剂制成乳化物注入Wistar大鼠皮内。观察大鼠实验性自身免疫性葡萄膜视网膜炎发作时间、临床表现及组织病理学改变,采用免疫组织化学S-P方法,检测EAU组和正常对照组视网膜Bcl-2、FasL的表达。结果EAU组动物的14只眼睛在免疫后不同时间发生临床可见的炎症(发病率为100%),正常对照组无临床可见炎症,所有具有阳性体征者均有组织病理学改变。免疫组织化学染色发现,EAU组及正常对照组视网膜组织中均有FasL表达,而EAU组Bcl-2表达增高,正常对照组Bcl-2低表达。结论Fas/FasL介导的细胞凋亡在维持免疫赦免中有重要意义;Bcl-2对Fas/FasL介导的细胞凋亡有调节作用;EAU的发生与细胞凋亡有关。Objective To detect the expression of FasL and Bcl-2 in experimental autoimmune uveoretinitis (EAU), and to investigate if apoptosis is involved in the pathogenesis of EAU. Methods S-Ag was extracted from fresh bovine retina, mixture of S-Ag and Freund's complete adjuvant (CFA) was injected intradermally into the footpads of Wistar rats to set up EAU animal models. FasL and Bcl-2 were examined in both EAU group and normal control group by immunohistochemistry. Results EAU was induced after immunization with S-Ag Wistar rats(The morbility was 100% ). High expressions of FasL were detected in EAU group and normal group. Bcl-2 was expressed in EAU group, but expressed at much lower level in that of normal group( P 〈 0.05). Conclusion Apoptosis mediated by Fas/FasL may play an important role in the maintenance of immune privilege .Bcl-2 may be involve and regulate the apoptosis of lymphocytes, which may be an important event in the pathogenesis of EAU.
