摘 要:目的探讨Fas和Fas配体(Fas/FasL)在人黑色素瘤中的表达及其与黑色素瘤恶性程度和细胞凋亡的关系。方法按不同病理分级,将标本分为恶性黑色素瘤(malignant melanoma,MM)伴淋巴结转移组(A组)、MM无转移组(B组)、良性黑素细胞痣组(C组)和良性黑素细胞痣旁正常组织对照组(D组),应用免疫组化SP法,检测Fas/FasL在4组标本中的表达情况;用DNA末端转移酶介导的缺口末端标记法(TUNEL)检测其细胞凋亡指数(AI)。结果Fas/FasL在不同组织中的表达强度有明显差异,Fas在A、B、C、D 4组中表达率分别为31.25%、57.89%、76.19%、92.85%(P<0.01),而FasL在上述组织中表达率分别为75.0%、52.63%、19.04%、3.57%(P<0.01)。AI比较:C组明显高于D组(t=3.054,P<0.01),B组高于D组(t=2.018,P<0.05),同时C组高于A组(t=2.051,P<0.05)。结论Fas/FasL表达情况反映黑色素瘤恶性程度,细胞凋亡的异常与黑色素瘤发展等过程有密切联系。Objective To study the expressions of Fas and Fas-ligand (Fas/FasL) in human melanoma, and its relations with malignant degree and apoptosis of human melanoma. Methods According to the pathology grade of melanoma tissues, specimens were divided into 4 groups: malignant melanoma (MM) with lymphoid node metastasis(group A), MM with no metastasis(group B), melanocytic nevus(group C), and normal control tissues closed to melanocytic nevus (group D). The expressions of Fas and FasL in different pathology grade of melanoma tissues were detected by immunohistochemistry SP method. And TdT-mediated. dUTP nick end labeling method (TUNEL) was used to analyze apoptosis index (AI) of tumor cell. Results The expressions of Fas/FasL were obviously different in different groups. The expression rates of Fas were 31.25%, 57.89%, 76.19%, and 92.85% in group A, B, C, and D, respectively ( P .〈 0.01 ). While the expression rates of FasL were 75.0 %, 52.63 %, 19.04 %, and 3 . 57 % in group A, B, C, and D, respectively ( P 〈 0.01 ). AI of group C was significantly higher than that of group D( t = 3.054, P 〈 0.01 ), AI of group B was higher than that of group D( t = 2.018, P 〈0.05) ,and AI of group C was higher than that of group A( t = 2.051, P 〈 0.05). Conclusion The expressions of Fas/FasL reflect the malignant degree of melanoma and there are closely association between the abnormality of tumor cell apoptosis and the development of melanoma.
