摘 要:目的研究N-乙酰半胱氨酸(NAC)对慢性重型肝炎肝衰竭时肝细胞增殖和生物转化功能的影响,并比较NAC和还原型谷胱甘肽(GSH)的疗效。方法体外慢性重型肝炎患者血浆培养C3A细胞,同时分别加入大、中、小三种剂量NAC及对应剂量的GSH,以正常血浆及培养基(10%FBS-MEM)为对照,分别于24 h、48 h、72 h三个时间点检测细胞安定代谢量的变化,并用MTT法测定C3A细胞活力。结果慢性重型肝炎血浆使细胞的安定代谢量降低,细胞增殖活力提高。NAC和GSH均可提高慢性重型肝炎血浆培养C3A细胞的安定代谢量,呈一定剂量依赖性,并可使细胞增殖活力进一步提高。结论NAC可以显著改善慢性重型肝炎肝衰竭时肝细胞增殖活力和生物转化功能,GSH也有相似功效,相同剂量的NAC优于GSH。Objective To investigate the effect of N-acetylcysteine(NAC) on hepatocyte proliferation and biotransformation function, and to compare the protective effect on hepatocytes between NAC and reduced glutathione(GSH) in vitro. Methods C3A cells were cultured with human chronic severe hepatitis plasma(CSHP), meanwhile, NAC and GSH were added in the media of C3A cells by 3 kinds of dosages(25 mmol/L, 5 mmol/L, 1 mmol/L). The C3A cells were cultured with normal human plasma or 10% FBS-MEM as control. We detected the metabolic rate of Diazepam and the cell activity by MTT at 24 h,48 h,and 72 h separately. Results CSHP.NAC and GSH promoted C3A cells proliferation,respectively. However, either NAC or GSH showed more strong pro-proliferation capacity than CSHP. CSHP decreased the metabolic rate of Diazepam of C3A cells. Both of NAC and GSH increased the metabolic rate of Diazepam of C3A cells. The effect of NAC and GSH were dose-dependent. Conclusion NAC can obviously promote hepatocyte proliferation and improve hepatocyte biotransformation function. GSH is similar to NAC in this kind of aspect, however is weaker comparing with GSH at the same dosage.
