摘  要:目的初步研究醋柳黄酮对大鼠非酒精性脂肪性肝病的疗效。方法雄性SD大鼠54只随机分组,正常对照组20只喂饲普通饲料,其余大鼠喂饲高脂饲料(88%标准普通饲料+10%猪油+2%胆固醇)。正常对照组和高脂饮食大鼠各10只于12周末处死观察造模效果。剩余高脂饮食大鼠24只分为模型对照组和醋柳黄酮治疗组。治疗组自13周开始进行醋柳黄酮灌胃治疗(10mg/d),于20周末处死各组大鼠,测定血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、γ-谷氨酰转肽酶(GGT)、碱性磷酸酶(ALP)、总蛋白(TP)、甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白(LDL)、高密度脂蛋白(HDL)、丙二醛(MDA)水平和超氧化物歧化酶(SOD)活性,同时观察肝组织病理学改变。结果20周末试验结束时,与正常对照组相比,模型组肝指数、血清ALT、AST、ALP、TP、TC、LDL水平显著升高(P≤0.01),肝组织脂肪变性、炎症及纤维化程度均较对照组重(P≤0.01)。醋柳黄酮治疗组血清ALT、AST、TC较模型对照组明显降低(P<0.05),ALP、GGT、TG等也有所下降,但差异无统计学意义;肝纤维化程度较模型组明显减轻(P≤0.01),而肝组织脂肪变及炎症程度较模型组有改善趋势,但差异无统计学意义。醋柳黄酮治疗组大鼠血清脂质过氧化产物MDA含量较模型组明显下降,差异有统计学意义(P<0.05),血清SOD活性较模型组有升高趋势,但差异无统计学意义。绪论醋柳黄酮对高脂饮食大鼠非酒精性脂肪性肝病有一定的治疗作用。Objective To investigate the primary therapeutic role of total flavones of Hippophae rhamnoides L (TFH)in treating nonalcoholic fatty liver disease(NAFLD)of rats. Methods Fifty-four male SD rats were randomly divided into 5 groups. The normal control groups (n = 20, N1 and N2 group) were fed with normal diet; NAFLD model groups (n = 22, M1 and M2 group) and therapeutic group(n = 12,T group)were fed with fat-rich diet, which was consisted with 10% lard oil and 2% cholesterol. Therapeutic group was treated with TFH at the dose of 10 mg/d from the 13^th week and finished at the 20^th week. The serum ALT, AST, GGT, ALP, TP, TG, TC, LDL, HDL, MOD levels and SOD activity were measured. And the degrees of hepatic histologic change were observed with HE and Masson stains. Results At the end of the experiment, compared with the normal control group, the liver index and the serum ALT,AST , ALP, TP, TC, LDL levels were increased significantly(P≤ 0. 01 ) in NAFLD model control group, and the hepatic steatosis, inflammation and fibrosis were more serious(P≤0.01 ). Compared with NAFLD model control group,the serum ALT,GGT, ALP, AST, TG,and TC levels were all down-regulated in the therapeutic group and ALT, AST, and TC level were decreased significantly(P≤0.05); The degrees of the hepatic steatosis and inflammation were improved with- out statistical significance while fibrosis degrees were lightened markedly(P≤0.01);The serum MOD was significantly decreased(P〈0.05) and the SOD activity in serum was up-regulated but with no statistical significance. Conclusion TFH has some therapeutic role in treating NAFLD of rats.
