摘 要:目的探讨核因子(NF)-κB环状哑铃型圈套寡核苷酸(CD—ODN)对膀胱癌NF—κB活性和细胞侵袭能力的影响以及有关的作用机制。方法脂质体介导法瞬时转染CD—ODN及其突变体(CD-ODN-mut)入膀胱癌BIU87细胞株,荧光素酶报告基因的方法检测不同处理组NF—κB活性的改变;逆转录.聚合酶链反应(RT-PCR)、Western—blot检测尿激酶型纤溶酶原激活剂(uPA)的表达水平;重建基底膜试验检测细胞侵袭能力的变化。结果转染CD-ODN后BIU87细胞株NF—κB活性被抑制,uPA的表达水平下降,细胞侵袭能力减弱,而转染CD—ODN—mut后无此作用。结论NF-κB组成性活化可促进uPA的表达,从而导致膀胱癌细胞的侵袭和转移。而CD—ODN能有效的阻断这一通路,降低膀胱癌的侵袭潜能。Objective To investigate the effect of circular dumbbell DNA oligodeoxynucleotides (CD-ODN) on NF-κB activity,cell invasion ability and the related molecule mechanism in bladder cancer cell line BIU87. Methods CD-ODN and CD-ODN-mut were transfected into human bladder cancer cell line BIU87 by Lipofectamine. Luciferase reporter was applied for detecting NF-κB DNA binding activity. The expression levels of uPA were detected by RT-PCR and Western blot, and the invasion ability by Transwell cell culture chamber. Results After transfection of CD-ODN into BIU87 cells, the activity of NF-κB was decreased significantly,the invasion ability weakened obviously and the uPA expression was inhibited as well. Conclusion NF-κB is constitutively activated in bladder cancer,which suggested that NF-κB activity is related to tumor progression due to its transcriptional regulation of uPA. CD-ODN can effectively inhibit the DNA binding activity of NF-κB in vitro and reduce the invasion ability of tumor.
