摘  要:目的:探讨碱性成纤维细胞生长因子(bFGF)对脑组织缺血再灌注神经元β-Catenin的调节作用及机制。方法:应用线栓法制作大鼠局灶性脑缺血再灌注模型,大脑中动脉阻塞1h再灌注损伤24h,采用TUNEL法、免疫组织化学SABC法及显微图像分析检测海马及顶叶皮质内神经元凋亡和β-Catenin的表达。结果:大鼠缺血再灌注海马及顶叶皮质内神经元凋亡数增加而β-Catenin免疫阳性反应产物较正常组增加,注射bFGF后神经元凋亡数减少,β-Catenin免疫阳性反应产物明显多于缺血再灌注组。结论:bFGF抑制缺血神经元凋亡,参与β-Catenin的调节,对缺血神经元有保护作用。Objective: To investigate the expression of β-catenin in hippocampus after cerebral ischemia and reperfusion in rats, and explore the regulative effect and mechanism of basic fibroblast growth factor (bFGF) to β-catenin on brain tissue. Methods: The model of middle cerebral arteries occlusion (MCAO) was performed with intraluminal filament method. The expression of β-catenin and apoptosis cell in the hippocampus and parietal cortex were detected with SABC immunohistochemical method and TUNEL method, and analyzed by microirnage system. Results: The expression of β-catenin in hippocampus and cortex was increased after cerebral ischemia and reperfusion; the expression of β-catenin in bFCJF group was increased, but cell apoptosis was decreased. Conclusion: The results indicate that bFGF depresses cell apoptosis, participating in the regulation of the expression of β-catenin in the ischemic neurons.
