摘 要:目的探讨低剂量化疗联合中药抗肿瘤疗效及Fas/FasL基因在其中的作用。方法将0、0.01、0.1、1μg/ml表阿霉素处理的肝癌细胞分别和有、无参芪处理的淋巴细胞共培养,观察低剂量化疗药联合中药或FasL单克隆抗体介入后抗肿瘤的效果;用PCR测定化疗药处理的肝癌细胞Fas/FasL基因的表达。结果1μg/ml表阿霉素处理的肝癌细胞OD值高于0、0.01、0.1μg/ml表阿霉素处理的肝癌细胞,未处理肝癌细胞的OD值最高;7402肝癌细胞表达Fas和FasL基因,与0μg/ml表阿霉素干预过的7402肝癌细胞FasL表达比较,0.01、0.1μg/ml的降低,1μg/ml的增高;FasL单克隆抗体介入后,0、0.01、0.1μg/ml表阿霉素处理的肝癌细胞OD值高于空白对照。结论低剂量化疗药联合中药抗肿瘤可减毒增效;Fas/FasL系统在抗肿瘤免疫机制中起重要作用。Objective To discuss the anti-tumor effect of low dose chemotherapy and traditional medicine, as well as the contribution of Fas/FasL gene. Methods In order to observe the anti-tumor effect of low dose chemotherapy and traditional medicine or that of monoclonal antibody intervention, lymphocytes treated by Shen Qi and those untreated were cocultivated with liver carcinoma cells treated by 0, 0.01,0.1, 1 μg/ml pharmorubicin respectively. The expression of liver cane.or cell gene Fas/FasL was detected by PCR. Results The OD value of liver carcinoma cells in 1 μg/ml group was higher than that in 0, 0.01 and 0.1 μg/ml groups, while the OD value of untreated liver carcinoma cells was the highest. Compared with the expression of 7042 liver carcinoma cell gene FasL treated by 0 μg/ml pharmoruhicin, the expression of 7042 liver carcinoma cell gene Fas/FasL was higher in 0.01, 0.1 μg/ml groups and lower in 1 μg/ml group. After the intervention of FasL monoclonal antibody, the OD value of liver carcinoma cells treated by 0, 0.01,0.1μg/ml phannorubicin was higher than the blank control group. Conclusion The combined anti-tumor therapy of low dose chemical drugs and traditional medicine can reduce the toxicity and enhance the effectiveness. The Fas/FasL system plays an important role in immunologic mechanism against tumor.
