摘 要:目的评价番茄种子经0.1%升汞和3%~4%次氯酸钠溶液灭菌的效果。方法将番茄种子浸于3%~4%次氯酸钠溶液中充分振荡20min,以灭菌水充分漂洗种子3~4次。另将番茄种子浸于75%酒精1~2min,弃酒精后加入足量的0.1%升汞溶液充分振荡10~15min,最后以灭菌水充分漂洗种子4~5次。将种子点播于TMT1固体培养基中。观察记录番茄种子经两种灭菌法后的最早出芽时间、子叶形成时间和全部出芽时间。结果经两种灭菌剂消毒后的番茄种子在最先出芽时间、子叶形成时间及全部出芽时间上均有高度显著性差异(P<0.01)。结论番茄种子消毒以3%~4%次氯酸钠溶液为佳。Objective To evaluate the efficiency of sterilizing tomato seeds with 0. 1% mercuric chloride and 3% -4% sodium hypochlorite solution. Methods Tomato seeds were firstly immersed in 3%-4% sodium hypochlorite solution for 20 min with shaking and then rinsed three or four times with sterile distilled water. Tomato seeds were firstly immersed in 0. 1% mercuric chloride for 10 to 15 min with shaking and then rinsed four or five times with sterile distilled water. Germinated the seeds on TMT1 medium. Observed the formost germinating time, the cotyledons emerging time and the whole germinating time of sterilization of tomato seeds with two sterilants respectively. Results The formost germinating time, the cotyledons emerging time and the whole germinating time of tomato seeds with two sterilants respectively had significant difference(P〈0. 01). Conclusion It's better for sterilizating tomato seeds with 3%-4% sodium hypochlorite solution.
