摘  要:目的研究澳洲茄胺盐酸盐(SBHL)对人宫颈癌细胞的增殖抑制作用和机制。方法用MTT法检测SBHL对ME180细胞增殖影响,用流式细胞技术和琼脂凝胶电泳法分析细胞凋亡、细胞周期变化,用电子显微镜观察细胞超微结构的变化。结果SBHL作用ME180细胞24h后,细胞存活率明显下降,而且随着澳洲茄胺盐酸盐浓度的增加,抑制作用增强(P〈0.01);SBHL可将ME180细胞周期阻滞于G1期,诱导细胞凋亡,细胞凋亡率明显增加(P〈0.01);细胞超微结构呈现典型的凋亡特征。结论SBHL可通过阻滞细胞周期发展发挥抑制ME180细胞增殖的作用,SBHL还可诱导细胞发生凋亡。Objective To study the inhibiting effects of SBHL on the growth of cervical cancer cells and investigate its mechanisms. Methods The proliferation of ME180 cells was observed by MTY assasy; The changes of cells cycles and apoptosis of ME180 cells treated with SBHL were analyzed by agar gel electrophoresis and FCM assay. The changes of the ultra structure of cells were observed by transmission eleconmicrograph. Results The proliferation of ME180 cells was inhibited after treating on SBHL for 24 h. The inhibiting effect increased gradually following the concentration of SBHL ( P 〈 0.01 ). The percentage of cells in G1 phase increased after the cells were treated with SBHL, SBHL arrested ME180 cells in G1 phase; apoptosis was induced and apoptosis ratio increased obviously( P 〈 0.01 ) ; typical apoptosis characteristic was present in the ultra structure of cells. Conclusion The inhibiting effects of SBHL on the growth of cervical cancer cells was carried out by development of cell cycles; apoptosis was induced by SBHL.
