摘  要:目的:研究黄芪(AS)对糖尿病大鼠心肌非酶糖基化反应和氧化应激反应的影响。方法:用链脲佐菌素腹腔注射制备糖尿病大鼠模型。黄芪注射液(10g/kg、8g/kg)ig治疗8周,分别测定各组大鼠体重、心脏质量指数、血糖、糖化血红蛋白、血清LDH、CK含量及心肌组织AGEs、MDA、NO含量及SOD、NOS活性。结果:8周实验终止时,糖尿病大鼠的心脏质量指数、血糖、糖化血红蛋白、血清LDH、CK和心肌AGEs、MDA、NO含量及NOS活性均明显高于正常组,而体重、心肌SOD活性明显低于正常组;黄芪治疗组的心脏质量指数、血糖、糖化血红蛋白、血清LDH、CK和心肌AGEs、MDA、NO含量及NOS活性均明显低于糖尿病组,但高于正常组,而体重、心肌SOD活性明显高于糖尿病组,但低于正常组。黄芪高、低剂量组间在血清LDH水平、心肌SOD活性及NO含量上有明显差异,高剂量组优于低剂量组。结论:黄芪能抑制糖尿病大鼠心肌非酶糖基化和氧化应激反应。Aim : To investigate the effects of Astragalus on nonenzymaticglycation and oxidative stress in diabetic rats. Methods: Forty male Wistar rats were randomly divided in to four groups: normal control group (NC) , diabetic group ( DM,ip 55mg/kg STZ) , Astragalus in high doses group ( AS2 , 10g/kg per day for 8 weeks. ) and Astragalus in low doses group ( AS1, 8g/kg per day for 8 weeks. ). At the end of 8 weeks ,the rats were killed. Body weight, heart mass/body weight, plasma glucose, glycosylated hemoglobin (HbA1c) , serum lactate dehydrogenase(LDH) and creatlne kinase(CK) were measured. Content of advancedglycationendproducts( AGEs), malondialdehyde (MDA) , nitric oxide (NO) and activity of superoxide dismutase (SOD) , nitric oxide synthase(NOS) in myocardial tissues were tested. Results: heart mass/body weight , plasma glucose , HbA1C , LDH and CK levels of serum , content of AGEs, MDA , NO and activity of NOS in myocardial tissues of diabetic rats were significandy higher than those of normal controls ( P 〈0.01 ). But body weight, SOD activity of heart tissue in diabetic rats were significandy lower than those of normal controls ( P 〈 0.01 ). After 8 weeks of treatment, heart mass/body weight , plasma glucose , HbA1 C , LDH and CK levels of serum ,content of AGEs, MDA , NO and activity of NOS in myocardial tissues of Astragalus groups were greatly lower than those of diabetic rats ,but higher than normal controls ( P 〈0.01 ,P 〈0.05). But body weight, SOD activity of heart tissue in Astragalus groups were significantly higher than those of diabetic rats , but lower than normal controls ( P 〈 0.01, P 〈 0.05 ). There is signiticandy difference in level of LDH, NO and activity of SOD between AS1 and AS2. Conclusion: Astragalus inhibits both nonenzymaticglycation and oxidative stress in heart of diabetic rats.
