可溶性大肠癌相关蛋白SCAP47的分离与鉴定

摘　　要：目的该研究通过差异显示技术确定大肠癌相关蛋白,以此制备相关抗体,构建了大肠癌细胞磁选分离系统。方法提取大肠癌及自身正常对照组织可溶性蛋白,行SDS-PAGE电泳,分析癌与对照组织的差异蛋白条带,找出出现率最高的癌上调差异条带,计算分子量,并观察该条带在非大肠癌标本中的出现率,富集该蛋白并鉴定纯度。结果在分子量14KD～66KD,存在2～5条不等的蛋白差异条带,或表现为癌组织较正常组织高丰度表达,或显示正常组织较癌组织高丰度表达。大肠癌组织高丰度蛋白条带中,42例中的27例(64.3%)癌组织出现在分子量47KD(SCAP47)左右条带处。18例腺瘤中有2例(11.1%)可见到47KD蛋白条带表达,12例溃疡性结肠炎和15例胃腺癌均未见到该蛋白条带。SCAP47的表达与大肠癌病理分期和病理分化程度无明显关系。结论该研究利用SDS-PAGE技术在配对的大肠癌标本中分离到的SCAP47蛋白显示与大肠癌组织有一定的特异性相关,提示其作为大肠腺癌相关肿瘤标志物用于大肠癌诊断的可行性。[Objective] To prepare relevant antibodies and construct magnet separating system for Colorectal cancer, according to the identification of colorectal cancer-associatel protein. [Methods] （1）All the soluble proteins from 42 cases with CRC and 42 normal colon tissues （control tissues） were extracted. （2）Samples of the soluble proteins were isolated by SDS-PAGE. （3）The electrophoresis images were analysed by Adobe Photoshop 6.0 software, and the molecular weight （MW） of the up-regulated proteins that occurred frequently in CRC tissue were calculated. （4）Enriched the target protein （soluble colorectal cancer associated protein, molecular weight 47 KD, SCAP47） by SDS-PAGE, and identified the purity. [Results] There were 2-5 differential protein fragments range of moleculhr weight from 14 KD to 66 KD among 42 pairs of colorectal cancer and their paracarcinoma tissues （normal tissues）. The up-regulated protein, MW 47 KD, appeared in 27 （27/42, 64.3%） CRC specimens, and 2 （2/18, 11.1%） colorectal adenoma samples, but absent in 12 ulcerative colitis and 15 gastric cancer specimens. There were no significant relativity among the appearance of SCAP47 protein and its pathologic stage and differentiation. [Conclusion] The findings support that the isolated SCAP47 protein may associate with genesis and developing of colorectal cancer.
