清热消痰散结方对S_(180)荷瘤鼠PGE-2IL-8表达的影响

摘　　要：目的:观察清热消痰散结方对热邪干预S180荷瘤鼠前列腺素E-2(PGE-2)及白细胞介素8(IL-8)的调节作用,探讨它对S180荷瘤鼠肿瘤的作用及作用机制。方法:利用腋下种植S180肉瘤细胞株建立肿瘤模型,并用灌胃乙醇的方法建立肿瘤热证模型,利用放射免疫方法检测S180小鼠血清中PGE-2的水平,采用双抗体夹心ABC-ELISA法测定热邪干预S180小鼠肿瘤组织中IL-8的含量。结果:成功建立S180肉瘤荷瘤鼠模型和热证S180肉瘤荷瘤鼠模型,病理组织学检测均为小鼠肉瘤。清热消痰散结方组、消痰散结方组、化疗组PGE-2、IL-8的含量均比热邪干预组PGE-2、IL-8的含量明显下调(P<0.05),热邪干预组比单纯肿瘤组PGE-2、IL-8水平明显升高,差异具有统计学意义(P<0.05)。结论:清热消痰散结方能抑制S180荷瘤鼠肿瘤的生长,其作用机制可能与下调S180小鼠肿瘤组织炎性因子PGE-2、IL-8的含量有关。Objective： To obersive the effect of PGE -2 ,IL -8 on S180 tumor mouse by interfere in hot pathogenic factor with QingRe XiaoTan Recipe, and to explore the mechanism of action to S180 tumor mouse. Methods： Establishing S180 tumor model, and establishing the model of pyretic syndrome by alcohol. The centrifuged blood was detected PGE - 2 by the method of radioimmunity, and observed the condition of hot syndrome. To detect IL - 8 in tumor tissue of S180 mouse be interfered in hot pathogenic factor by method of ABC - ELISA. Results： The the tumor model of hot syndrome was sueeessful to be established. The contents of PGE - 2, IL - 8 of QingRe XiaoTan group, XiaoTan group were lower than ReXie groups, the difference was statistics significance（ P 〈 0.05 ） , and the contents of PGE - 2 ,IL - 8 of ReXie group was higher than tumor group, the difference was statistics significance（ P 〈0.05）. Conclusions ：It can inhibit the growth of S180 tumor by WenZhong XiaoTan Recipe. The mechanism may be down regulation the level of inflammatory factor PGE - 2, IL - 8 on S180 tumor mouse.
