摘 要:目的研究在不同CO2气腹压及不同时段下正常及缺氧缺血性脑损伤大鼠脑组织S-100mRNA(S-100)、神经元特异性烯醇化酶(NSE)mRNA表达的变化,并进而探讨CO2气腹对中枢神经系统的影响.方法采用健康成年Wistar大鼠建立缺氧缺血性脑损伤模型和气腹模型,用RT-PCR技术检测脑组织中S-100、NSEmRNA表达的变化.结果(1)CO2气腹后,S-100mRNA表达在A5-1组(64.215±10.404)和A10-2组(67.020±13.339),NSE分别为(8.976±2.379)和(9.183±3.365),对照组S-100、NSEmRNA分别为(59.340±11.378)和(8.500±2.940),S-100、NSEmRNA表达在CO2气腹后逐渐增加,但与对照组比较,差异无统计学意义(P>0.05);(2)S-100、NSEmRNA表达在B组较对照组明显升高(P<0.05),在B10-2组与B组比较有统计学意义(P<0.05).结论在10mmHg、2h范围,CO2气腹对正常大鼠脑组织S-100、NSEmRNA表达无明显影响;对缺氧缺血性脑损伤大鼠有显著性影响,应尽可能缩短气腹时间及采取相应的调控措施.Objective To evaluate the changes of expressions of the mRNA of S-100 protein (S-100) and neuron specific enolse (NSE) at different period and pressure of carbon dioxide pneumoperitoneun (CO2 PP) and explore the effects of CO2 PP on central nervous system (CNS) in normal and hypoxic-ischemic brain damage rats. Methods Healthy adult Wistar rats were used to establish the models of CO2 PP and of hypoxic-ischemic brain damage. By using the RT-PCR technique, the expressions of mRNA of S-100 and NSE in the brain tissue were tested. Results (1) After CO2 PP, the expressions ofmRNA of S-100 and NSE were (64.22±10.40), (8.98 ±2.38) respectively in A5-1 group, (67.02 ± 13.34), (9.18 ±3.37) respectively in A10-2 group, and (59.34 ±11.38),(8.50 ± 2.94) respectively in control group. The expressions of S-100 and NSE increased with the increase of pressure and time of CO2 PP. There were no significant difference between them (P 〉 0.05 ). (2) The expressions of mRNA for S-100 and NSE increased progressively in B group, B5-1 group and B10-2 group than those in control group. There were significant difference between B group and control group (P 〈 0.05) Or Bi0-2 group and B group (P〈0.05). Conclusions Hours of CO2 PP at pressure of 10 mmHg and within 2 hours has no significant effect on the expressions of the mRNA of S-100 and NSE in normal rat, but significant on the expressions of the mRNA of S-100 and NSE in hypoxic-ischemic brain damage rat. It is advisable that PP time be shortened or control measures be taken.
