摘  要:目的检测膀胱移行细胞癌(BTCC)组织中缺氧诱导因子1α(HIF-1α)和第10号染色体丢失的张力蛋白同源的磷酸酶基因(PTEN)的表达情况,探讨两者在BTCC中表达的可能临床意义。方法应用免疫组织化学检测BTCC组织中HIF-1α和PTEN的蛋白表达,分析其表达与肿瘤部分临床病理特点的关系,并分析两者表达的相关性。结果BTCC组织中HIF-1α阳性表达率为52.63%(30/57),其表达与肿瘤的临床分期及复发明显相关;BTCC组织中PTEN阳性表达率63.16%,随着肿瘤分级、分期的增加PTEN表达显著降低(P<0.05)。而且,HIF-1α与PTEN的表达具有明显负相关性(γ=-0.360,P=0.006)。结论HIF-1α及PTEN在BTCC的侵袭等生物学过程中具有重要的作用,HIF-1α和PTEN可能可以用于临床监控BTCC的进展。Objective To investigate the clinicopathological significance of HIF-1α and PTEN expression in bladder transitional cell carcinoma(BTCC). Methods Hypoxia inducible factor 1 α (HIF-1α) and phosphatase and tensin homology deleted on chromosome ten (PTEN)protein expression were determined using immunohistochemistry. The relationship between HIF-1α,PTEN expression and various clinicopathological parameters including pathological grade and clinical stage was analyzed and the correlation between HIF-1 α expression and PTEN expression was also analyzed. Results Over-expression of HIF-1 α was frequently (30 of 57 cases,52.63% ) detected in BTCC tissues. HIF-1α expression was significantly associated with invasive bladder cancer. The positive expression rate of PTEN in BTCC was 63.16%. Significant differences existed in the expression of PTEN in carcinomas form different pothological grades and clinical stages ( P 〈 0.05 ). There was significant negative correlation between the expression of HIF-1 α protein and the PTEN protein expression ( γ= - 0. 360, P = 0.006). Conclusion HIF-1α and PTEN may play a critical role in the invasion of bladder carcinoma and may be used to estimate the progression of bladder carcinoma.
