摘 要:目的研究苦参碱对人鼻咽癌细胞系CNE1-GL增殖及转移相关能力的影响及其作用机制。方法以MTT法检测苦参碱对人鼻咽癌CNE1-GL细胞增殖及对细胞外基质黏附能力的影响;Transwell小室法检测其对CNE1-GL细胞侵袭重组人工基膜能力的影响;免疫细胞化学方法检测药物作用前后CNE1-GL细胞EBV-LMP1蛋白的表达。结果苦参碱对CNE1-GL细胞的增殖及对细胞外基质的黏附能力有明显的抑制作用,该抑制效应随苦参碱浓度增加和作用时间延长而增加;1.5g/L苦参碱能明显抑制CNE1-GL细胞体外侵袭人工基膜能力,抑制率达(42.51±11.06)%,作用于CNE1-GL细胞24h后EBV-LMP1蛋白的表达明显下调。结论苦参碱能抑制CNE1-GL细胞的增殖能力和转移能力,其抗肿瘤转移的机制与EBV-LMP1蛋白表达的改变有关。Objective To investigate the effect of matrine on the proliferation and the metastatic abilities of the nasopharyngeal carcinoma CNE1-GL cells and its involved mechanism. Methods The MTT assay was used to examine the effect of matrine on the proliferation of CNEI-GL cells and its adhesion to extracellular matrix matrigel;Transwell chamber assay was performed to determine the effect of matrine on the invasion of the cells ;the expression of EBV-LMP1 protein in CNE1-C,L was detected by imnmnocytochemical SP methods. Results Matrine inhibited CNE1-GL cell proliferation and adhesion to the matrigel significantly, and this effect augmented with the up-regulation of the dose and extension of the treatment time ; 1.5 g/L Matrine inhibited CNE 1-GL cells invasion to the recombinated basal membrane significantly, with the IR, (42.51 ± 11.06)% , and it down-regulated the expression of EBV-LMP1 significantly after 24 hours. Conclusion Matrine inhibits the proliferation and metastatic potentials of CNE1-GL cells in vitro,possibly involved with the down-regulation of EBV-LMP1 protein.
