大鼠肝癌发生过程中PTEN蛋白在卵圆细胞内的表达及意义

摘　　要：目的研究PTEN蛋白在大鼠肝癌发生过程中的表达与肝癌的关系。方法将60只SD大鼠随机分为实验组和对照组。用3’-甲基-4-二甲基氨基偶氮苯(3’-Me-DAB)诱癌,再分别用RT-PCR和Westernblot检测诱癌4～20周的大鼠卵圆细胞中AFPmRNA和PTEN蛋白的表达。结果PTEN表达的下调与癌症的发生过程相关,AFPmRNA的表达相对稳定。结论PTEN在肝癌发生过程中的表达逐渐降低可能与卵圆细胞的癌变相关。Objective To analyze the expression of phosphatase and tensin homolog deleted on chromosome ten（PTEN）and its association with hepatocellular carcinogenesis in rats. Methods Sixty SD rats were divided into test group and control group randomly. The hepato-carcinogenesis was induced by 3＇-methyl-4-dimethylaminoa-zobenzene（3＇-Me-DAB）. The AFP mRNA and PTEN protein expression in oval cells was detected by RT-PCR and Western blot every 2 weeks from the 4th to the 20th weeks. Results Down-regulation of the PTEN protein expression in the liver of murine model was related with hepatocellular carcinogenesis. AFP mRNA was expressed constantly during the carcinogenesis. Conclusion The gradual reduction of PTEN protein in the carcinogenesis was probably related to the carcinogenesis of oval cells.
