摘 要:采用GS10A型逆流色谱仪和AKTAprime plus层析系统共同组成的高速逆流色谱联用系统对藤茶提取物中的二氢杨梅素和杨梅素进行了同时纯化。选用溶剂系统为石油醚/L酸乙酯/甲醇/水/三氯乙酸(体积比1:3:2:2:0.05),在逆流色谱仪转速为700~800r/min,进样体积为2~50mL、流速为1.5~2.5mL/min的条件下,在5h内能同时将粗提物中的二氢杨梅素和杨梅素提纯到99%以上,50g粗藤茶样品连续分离其分离得率分别为85.5%和87.6%。重复使用溶剂对分离效果无明显影响。A combined AKTA-HSCCC system( GS10A-Countercurrent Chromatography and AKTAprime pins detection system)is used to purify dihydromyrlcetin and myrlcetin from extract of Am,pelopsis grossedentata in this paper.The results show that, with a solvent system composed of mineral ether/ethyl acetate/methanol/water/ trichloroacetic acid ( 1 : 3 : 3 : 2 : 0.05, v/v), dihydromyricetin and myricetin from extract of Am,pelopsis grossedentata can be purified into over 99% purity at the same time. Under the separation conditions determined as 700 to 800 r/rain of rotation rate,2 to 50 mL of injection volume, 1.5 to 2.5 rnL/min of flow rate and 5 h of sepertation time,26.5 g dihydromyricetin and 9.6 g myricetin can be obtained at a high purifty of over 99%, with separation rate of 85.5% and 87.6%, respectively, from 50 g extract of Am,pelopsis grossedentata. Reusing the solvent has no obvious negative effect in the separation process.
