摘  要:目的研究滤蓝光人工晶状体对猪视网膜色素上皮(RPE)细胞的保护作用。方法原代培养猪RPE细胞,3~5代用于实验,设滤蓝光人工晶状体组,普通晶状体组,直接暴露组和非暴露组。4组均在光照强度为(50000±10000)lux的蓝光照射2h后进行彗星实验,在荧光显微镜下观察,计数拖尾细胞的尾长,尾相,以评价RPE细胞的DNA损伤情况。结果直接暴露组和非暴露组比较平均尾长(MTL)、平均尾相(MTM)差异有统计学意义(P<0·01),普通晶状体组细胞损伤小于直接暴露组但差异无统计学意义(P>0·05),滤蓝光人工晶状体组与直接暴露组相比较MTL和MTM差异有统计学意义(P<0·01),滤蓝光人工晶状体组与普通人工晶状体组比较MTL和MTM差异有统计学意义(P<0·01)。结论蓝光可以造成猪RPE的DNA明显损伤,滤蓝光人工晶状体对猪RPE细胞的蓝光损伤保护作用明显优于普通人工晶状体。Objective Retinal pigment epithelium(RPE) cells was an important cell,and its disfunction could result in a series of retinal disease. The damage of RPE can be induced by light in the blue region of the spectrum. Present study was carried out for the aim of exploring the protective effect of blue-light absorbing intraocular lens on retinal pigment epithelium(RPE) cells. Methods RPE cells of pig were primarily cultured and passaged in F12 medium containing 10% fetal bovine serum(FBS). The third to fifth generation of cells were used and divided into 4 groups for the conduction of the experiment. The cultured pig RPE cells were exposed to the blue light of 50 000 ± 10 000 lux for 2 hours in the groupⅠ. The conventional lens and blue-light absorbing lens was put in the front of light source respectively in group Ⅱ and Ⅲ. Cultured cells did not irradiated in group Ⅳ. The diameter,mean tail long (MTL) and mean tail moment(MTM) of RPE cells were analyzed by comet test to detect the DNA damage of cells. Results Obvious comet-like tail of cells was found in group Ⅰ and Ⅱ ,and a small tail was seen in group Ⅲ Only spot-like fluorescence tail was displayed in group Ⅳ. There was a significant difference in MTL and MTM between blue light exposed cells(group Ⅰ )and unexposed cells(group Ⅳ ) (P 〈 0. 01 ). However, no significant difference was found in MTL and MTM between the cells protected by conventional lens(group Ⅱ ) and blue light exposed cells (group Ⅰ ) (P 〉 0. 05). And a significant difference was exhibited in MTL and MTM between the cells protected by blue-light absorbing lens ( group Ⅲ) and blue light exposed cells ( group Ⅰ ) (P 〈 0. 01 ). Meanwhile, there was a significant difference in MTL and MTM between the cells protected by blue-light absorbing lens (group Ⅲ)and the cells protected by conventional lens( group Ⅱ)( P 〈 0.01 ).
