摘 要:目的观察人视网膜发育和分化过程中基质金属蛋白酶9(MMP-9)的表达水平与细胞分布特点,探讨其在人视网膜发育和分化过程中的可能作用。方法胚胎发育早期、中晚期及成人视网膜石蜡切片,抗MMP-9免疫组织化学染色,观察MMP-9表达水平和分布的变化。结果胚胎发育早期的视网膜仅在外成神经细胞层的外侧可见弱阳性细胞;胚胎发育中期外成神经细胞层的外侧,即将来要发育成盘膜处出现明显的特异性染色,而其他细胞染色仍然较弱;成人视网膜的外核层出现显著的特异性染色细胞,胞质明显着色,胞体轮廓清晰,并可观察到锥体的结构以及突触的特异染色,判断这些细胞主要是光感受器细胞中的视锥细胞。结论MMP-9在视网膜中的表达与视网膜的成熟和细胞分化关系密切,在光感受器中的视锥细胞高水平表达,提示MMP-9可能参与视觉信号的处理。Objective The matrix metalloproteinases(MMPs) have been identified to involve in many pathologic processes such as tissue repairing or remodeling, inflammation, tumor metastasis, etc.. To better understand the role of MMPs in fundus diseases,we examined the expression and location characteristics of MMP-9 during development and differentiation of human retina. Methods Four 15- and 16-week-old,three 22- and 26 week-old fetal and four 20- and 40-year-old adult eyes from donor were fixed in ethanol-glacial acetic acid and embedded in paraffin. The expression and cellular distribution of MMP-9 in the human retina were investigated by immunohistochemistry assay in the retina of the early stage, mid-late fetal stages and the adulthood stage. The structure of retinas was observed and analyzed under the light microscope. Results In the retina of the early fetal stage, MMP-9 protein appeared with the relatively weaker staining intensity in the outer neuroblast layer. In the mid-late fetal stage,MMP-9 immunoreactivity was mainly restricted to profiles in the outer neuroblast layer which would develop into the outer segment of photoreceptor. However,in the retina of adult stage, specific immunoreactivity signal appeared in the soma and processes of cells characterized by cone photoreceptor according to the structure and the anatomical distribution of these outer nuclear cells. Conclusion Expression pattern of MMP-9 varies significantly as the development of human retina. It is expressed mainly in the cone photoreceptor, indicating that MMP-9 is probably involved in the formation or function of photoreceptor. These outcomes offer a basis for further understanding the role of MMP-9.
