摘 要:目的研究大鼠视网膜缺血再灌注(RIR)组织中生存素survivin和caspase-3蛋白动态表达的生物学意义。方法选用SD大鼠42只,随机分为两组:A组为对照组(6只);B组为实验组(每个时间段6只,共6个时间段)。B组采用结扎单侧颈总动脉方法诱导大鼠视网膜缺血60min,恢复视网膜血供,建立RIR模型。缺血60min,再灌注1、6、12、24、48、72h。A组暴露单侧颈总动脉不结扎。按规定时间段(1、6、12、24、48、72h,)处死动物,取眼球切片行苏木精-伊红染色,survivin原位杂交,caspase-3免疫组织化学观察。结果实验组survivin表达延迟于caspase-3,再灌注12h开始增加,24h达高峰,以后逐渐降低,与对照组相比差异有统计学意义(P<0·01),至72hsurvivin表达水平比对照组略微增高。结论Survivin与caspase蛋白参与大鼠缺血再灌注眼组织细胞的凋亡过程。Objective Retinal ischemia-reperfusion injury involves in many retinal diseases. Present study was to investigate the biologic significance of the dynamic expressions of survivin and caspase-3 in the models of rat retinal ischemiareperfusion(RIR) injury. Methods Fourty-two SD rats were divided into control group(6 rats) and experimental group( RIR group,36 rats) randomly,and RIR group was subdivided into 1 hour,6,12,24,48 and 72 hours groups according to different time after RIR injury. The rat single common carotid artery was exposed to air without ligating in the control group, and rat single common carotid artery was ligated for an hour in group B to establish the model of rat retinal RIR injury. The apoptosis in retina was detected by TUNEL assay under the light microscope. Survivin was measured by the in situ hibridization ,and caspase-3 was measured by the immunohistochemistry. Results Caspase-3 was mainly expressed in cytoplasm in retinal ganglion cell layer, inner plexiform layer and inner nuclear layer,exhibiting the brown staining. In experimental group,the expression of caspase-3 was presented in 1 hour after RIR and peaked in 6 hours and decreased 12 hours later gradually. There was a statistically significant difference between experimental group and control group in the expression of caspase-3 at various time points ( P 〈 0.05 ). The expression of survivin was gradually increased at 12 hours after RIR and peaked in 24 hours after RIR, showing a remarkable difference between control group and experimental group( P 〈 0. 01 ). At the 72 hours after RIR, the expressive level of survivin showed an insignificant increase in comparison with control group (P 〉 0.05 ). Conclusion Survivin and caspase-3 are involved in the procedure of cellular apoptosis in the RIR eye in rat.
