摘  要:目的研究羟基红花黄色素A(hydroxysafflor yellow A,HSYA)对晶状体氧化损伤的保护作用。方法取正常兔培养的透明晶状体分为4组:对照组、Feton模型组、2.40mmol·L-1HSYA组和6.24mmol·L-1HSYA组进行体外培养,培养24h后观察各组晶状体的混浊情况;利用光镜和电镜观察组织学变化;利用分光光度计测各组SOD、GSH-Px及MDA组织化学变化。结果(1)Feton模型组晶状体全部混浊,对照组、2.40mmol·L-1HSYA和6.24mmol·L-1HSYA组分别有17%、75%和33%发生雾状混浊;(2)光镜和电镜下见对照组组织结构基本正常,Feton模型组严重损害,2.40mmol·L-1HSYA组和6.24mmol·L-1HSYA组轻微损害;(3)4组经方差分析,差异有统计学意义(P<0.05)。经多重比较:和对照组相比,Feton模型组2种抗氧化酶水平显著降低(P<0.01),MDA水平显著升高(P<0.01);和Feton模型组相比,2.40mmol·L-1和6.24mmol·L-1HSYA组上述指标有不同程度改善,且呈剂量依赖性。结论HSYA可在体外保护晶状体免受氧化损伤,阻止脂质过氧化过程和白内障的形成,且高浓度组更为有效。Objective To evaluate the effect of hydroxysafflor yellow A(HSYA) on protecting lens against oxidative stress.Methods Rabbit lenses cultured in vitro were divided into four groups,inlcuding control group,Feton model(oxidative stress) group,2.40 mmol·L-1 HSYA group and 6.24 mmol·L-1 HSYA treatment group.After 24 hours,the transparency of all lenses were examined,the histological changes of all lenses were observed by light and electron microscopies,and the levels of SOD、GSH-Px and MDA were measured,respectively.Results After incubated for 24 hours,all lenses of Feton model group developed into cataract.And the ratio of lenses that were showed swelling in control group,2.40 mmol·L-1 HSYA group,6.24 mmol·L-1 HSYA group were 17%,75%,33%correspondingly.The structure of lenses in control group was norma1,but was damaged severely in Feton mode1.Slight damage was found in lenses which were cultured by 2.40 mmol·L-1 HSYA and 6.24 mmol·L-1 HSYA media.The result had statistical significance by variance analysis of four groups(P〈0.05).Compared with control group,the levels of two antioxidase of Feton group were decreased significantly (P〈0.01),whenas the level of MDA was increased significantly (P〈0.01);Compared with Feton group,all indexs above in 2.40 mmol·L-1 HSYA and 6.24 mmol·L-1 HSYA treatment groups had been improved to different degree and had dose dependence.Conclusion HSYA can protect lens against oxidative stress by preventing it from lipid peroxidation and cataract formation in vitro,and the 6.24 mmol·L-1 HSYA group may be more effective.
