摘  要:目的运用描述性毒理学初步探讨汉防己甲素(tetrandrine,Tet)对兔眼的毒性作用。方法健康新西兰大耳白兔30只,随机分为4组,1g·L-1组、3g·L-1组、9g·L-1组及正常对照组,模拟临床连续用药3个月。进行急性眼刺激性实验,临床裂隙灯观察,A超测量角膜厚度,考马斯亮兰测房水蛋白浓度,角膜内皮细胞茜素红台盼兰联合染色及组织病理学和透射电镜检查。结果1g·L-1、3g·L-1Tet对眼均无刺激性;9g·L-1Tet有轻度刺激性。临床裂隙灯观察,1g·L-1组眼前节正常,3g·L-1、9g·L-1组出现不同程度角膜水肿,9g·L-1组出现明显炎症反应。1g·L-1组与正常对照组比较,角膜厚度、房水蛋白含量、角膜内皮细胞活性率均无显著性差异(P>0.05);3g·L-1、9g·L-1组与正常对照组比较,角膜厚度、房水蛋白含量显著增加,内皮细胞活性率显著降低(P<0.05)。组织病理及超微结构检查示:1g·L-1组无病理及超微结构改变;3g·L-1组可见角膜水肿,睫状体上皮水肿,晶状体上皮细胞胞核出现核沟;9g·L-1组可见角膜上皮细胞、角膜细胞和内皮细胞水肿、变性、坏死,虹膜和睫状体上皮细胞水肿,晶状体上皮细胞胞质出现半透明空化区,线粒体肿胀。结论Tet的浓度、用药时间与刺激性及毒性呈正相关,1g·L-1浓度Tet对眼组织无刺激性、毒性,应可安全应用于眼部。Objective To investigate the eye toxicity of tetrandrine (Tet) with descriptive toxicology.Methods Thirty healthy New Zealand white rabbits were randomly divided into normal group,1 g·L-1 group,3 g·L-1 group and 9 g·L-1 group,giving Tet for 3 months.Acute eye irritation experiment,slit lamp microscope examination,the thickness of cornea with A ultrasound,the concentration of protein in the aqueous humor,the percentage of survival corneal endothelial cells,histopathology and electron microscope examination was taken.Results 1 g·L-1 and 3 g·L-1 Tet had no irritation,while 9 g·L-1 group had mild irritation;The results of slit lamp microscope examination were normal in 1 g·L-1 group,while 3 g·L-1 and 9 g·L-1 group caused different degree cornea edema,and 9 g·L-1 group caused inflammation reaction;There was no significant difference in thickness of cornea,the content of protein in aqueous humor,the percentage of survival corneal endothelial cells between 1 g·L-1 and normal group(P〉0.05),but there was significant difference between 3 g·L-1,9 g·L-1 and normal group(P〈0.05);Histopathology and electron microscope examination were normal in 1 g·L-1 group,but 3 g·L-1 with corneal edema,ciliary epithelium edema and groove in lens epithelium nucleus,and 9 g·L-1 group with edema,degeneration and apoptosis of corneal epithelium,corneal cells and epithelium.Conclusion The concentrations of Tet and medication time have positive relation with irritation and toxicity.1 g·L-1 Tet and concentration under 1 g·L-1Tet can be applied on ocular safely,but further studies on the security of Tet applied to human eyes need to taken.
