摘 要:以4个不同的水稻品种为材料,分别采用6种不同的单一组分和复合组分的醇溶蛋白提取剂,在同一条件下提取水稻种子的醇溶蛋白。通过比较SDS-PAGE电泳图谱发现:单一组分提取剂中100%乙二醇效果最好,用此提取剂提取的醇溶蛋白经SDS-PAGE电泳后,图谱中带型清晰,分辨率高,条带位置准确,是水稻醇溶蛋白理想的提取剂;5%2-巯基乙醇提取效果最差。此外,还用得出的最佳提取剂提取了另外36个水稻品系的醇溶蛋白,用考马斯亮蓝G-250染色法和SDS-PAGE比较分析了醇溶蛋白的总含量和组成的多态性。结果表明:不同水稻品系在醇溶蛋白总量、各组分的百分含量等上表现出一定的多态性。The rice prolamines were extracted from the seeds of 4 lines with 6 different extracting solvents on the same conditions. The results showed that the extraction of prolamine was most efficient with the extracting solvent 100 % ethylene glycol. Using this extractant, suitable prolamine bands could be resolved by SDS-PAGE, and its electrophoregram was clarity. Therefore, it was the best extractant for rice prolamines. In addition, polymorphisms of rice prolamine extracted by the best extractant were analyzed. The results showed that there were some polymorphisms in the total prolamine quantities, composition and percentage among the lines studied. Electrophoregrams of the extracted prolamines could be used to analyze the genetic divemity and phylogenesis of rice populations.
