摘 要:目的探讨纳米硫硒化镉(CdSeS)对细胞DNA损伤作用。方法用不同剂量的纳米CdSeS(0、0.1、0.2、0.4 mg/ml)经小鼠尾部静脉注射,一次性染毒,3 d后通过彗星实验检测肝细胞和脑细胞的核DNA损伤。结果在纳米CdSeS低浓度(0.1 mg/ml)下,尾部DNA百分率(Tail DNA%)和尾矩(Tail Moment)与对照组相比无显著差异性,在高浓度(0.2、0.4 mg/ml)下,各染毒组细胞尾部DNA百分率和尾矩显著增加(P<0.01)。结论纳米CdSeS能通过血脑屏障,导致小鼠脑细胞DNA损伤,并能导致小鼠肝细胞DNA损伤。并且,脑和肝的细胞核DNA损伤趋势一致,但是在同一浓度染毒组,脑细胞核DNA的损伤较肝小。Objective To explore the DNA damage effects on cells induced by nanosized CdSeS. Methods Using different concentrations of CdSeS nanometer (0,0.1,0.2,0.4 mg/ml) to do one-time injection at the vein of mice tail, and test the DNA damage of brain cells and liver cells through the comet experiment. Results CdSeS nanometer did not cause DNA damage at low levels (0.1 mg/ml). However,tail DNA% and tail moment were both significantly increased in the high concentrations (0.2, 0.4 mg/ml) of nanosized CdSeS as compared with the control group (P〈 0.01). Conclusion The nano-CdSeS can pass through the blood-brain barrier and lead to DNA damage in mice brain cells, and also lead to DNA damage in the liver ceils, In addition, the DNA damage of nucleolus in brain and liver tends to be the same. But at the same concentration of the affected group, the DNA damage in brain cells is weaker than that in the liver cells.
