摘 要:目的探讨全反式维A酸对体外培养皮肤鳞癌细胞增殖的影响。方法传代培养皮肤鳞癌细胞,不同浓度维A酸作用后,MTT法观察维A酸对细胞增殖的影响,流式细胞仪进行细胞凋亡及周期分析。结果低浓度维A酸作用后细胞增殖率高于对照组,其中1μM浓度组作用最强,而高浓度维A酸作用后细胞增殖率低于对照组,且呈剂量依赖性抑制;ATRA不能诱导A431细胞发生显著凋亡;维A酸对皮肤鳞癌细胞的促进增殖作用可能与S期细胞数百分比增多有关。结论全反式维A酸对体外皮肤鳞癌细胞的增殖抑制作用有严格的剂量范围,为皮肤鳞癌的临床治疗提供实验依据。Objective To investigate the effect of all - trans retinoic acid(ATRA) on the proliferation of human epidermoid carcinoma ( A431 ) cells. Methods MTT assay was used to determine the effects of all-trans retinoid on human epidermoid carcinoma A431, Microscopy was used to observe cell morphological changes. Flow eytometry was performed to investigate the influence of all - trans retinoid on cell cycle. Results The proliferation rate of cells was higher in the low-dose retinoie acid group, especially in 1 μmol/L group. The percent S-phase ( by flow cytometry) in 24-hour ATRA-treated cells was higher than that in their control vehicle-treated cells in A431 cell lines, Conclusion The anti-proliferation effect of all-trans retinoic on cell proliferation of human epidermoid carcinoma A431 seems to be achieved only in the presence of a critical ATRA concentration.
