摘  要:目的探讨门静脉来源内毒素在肝脏代谢过程中,内毒素在肝脏细胞内的时相性分布规律.方法 Wistar 大鼠门静脉内注入内毒素10μg/100g 体重,0,2,5,10,30,60分钟后取肝脏固定,应用单克隆抗内毒素抗体,采用免疫荧光及共焦点激光扫描技术,对内毒素在肝脏细胞内的时相性分布进行了观察.结果内毒素注入2分钟后,肝窦壁细胞内抗内毒素荧光活性增高,10分钟后达高峰,这些阳性肝窦壁细胞分布特性与枯否细胞分布相符.同时,内毒素注入2分钟后肝细胞内抗内毒素荧光活性增强,30分钟后呈高峰.肝内胆管上皮细胞亦出现强烈抗内毒素荧光反应,半小时达高峰.这些阳性肝脏细胞抗内毒素荧光活性均局限于胞浆,呈小片状或点状分布。结论门静脉来源内毒素在肝脏内迅速被细胞所摄取,且枯否细胞,肝细胞及肝内胆管上皮细胞均参与这一降解过程.Objectives To reveal the detoxicative metabolism and cellular distribution of endotoxins from the portal vein in the liver in rats.Methods Lipopolysaccharide(LPS)at a dose of 10μg/100g body weigh was injected to Wistar rats by the portal vein.Zero,2,5,10,30,60 minutes after LPS injection,liver Samples were obtained and cellular distribution in the was observed by the immunofluorescent method using a monoclonal antibody specific to LPS.Results Two minutes after the injection of LPS,LPS were first detected in both Kupffer cells and hepatocytes,and were showed a peak at 10 min and 30 min respectively.The increase of fluorescent intensity to LPS was also found in intrahepatic bile ductular epithelial cells of small and large bile ducts 10 min after LPS injection.The fluorescence internsity against LPS was found only in the cytoplasma of these cells.Conclusinos In the liver,LPS from the portal vein is quickly taken up by liver cells,and besides Kupffer cells,hepatocytes and bile ductular epithelial cells are also involved in the detoxication LPS in the liver.
