摘 要:目的探讨肿瘤细胞膜组成及流动性在癌转移中的作用及机制。方法以具有不同转移表型的人肺腺癌细胞系Anip及AGZY为对象,采用32P标记法及薄膜层层析和气相层析技术分析两个细胞系的膜脂及脂肪酸组成,并用荧光偏振技术测定两者的膜脂流动性。结果两者在磷脂和脂肪酸及膜流动性上均有显著不同,转移性较高的Anip细胞质膜含有较多的磷脂酸(PA)和磷脂酰肌醇(PI)及较少的肉豆蔻酸(myristicacid),并具有较低的流动性。结论癌细胞的转移能力与膜脂肪酸组成有密切关系。PURPOSE To study the relationship between tumor cell membrane composition and metastatic ability. METHODS The membrane lipid composition and fatty acid composition of two human lung adenocarcinoma cell lines with different metastatic ability,the high metastatic cell line Anip and the low metastatic cell line AGZY were studied by thin layer chromatography and gas chromatography method. The fluidity of these cell lines was also determined by flurorescence polarization technique using DPH(1,6 diphenyl 1,3,5 hexatriene) as probe. RESULTS The high metastatic cell line Anip has more phosphatidic acid(PA) and phosphatidylinositol(PI) in membrane lipid composiition than the low metastatic cell line AGZY,but has less myristic acid in fatty acid composition and lower degree of membrane fluidity. CONCLUSIONS It is suggested that metastatic ability of human lung adenocarcinoma may be associated with the alteration of lipid and fatty acid composition.
