摘 要:目的:探讨人工合成E-选择素对大鼠脑缺血再灌注损伤的保护作用机制。方法:雄性SD大鼠90只,随机分为3组:①假手术组;②缺血再灌注组;③人工合成E-选择素治疗组(E-选择素治疗组)。大鼠局灶性脑缺血再灌注模型中E-选择素治疗组建立模型前5min从股静脉注入人工合成E-选择素10mg·kg-1。在不同时间点(缺血再灌注后2、6、12、24、48和72h)用ELISA法测定血浆IL-1β、TNF-α含量。分别在光镜和电镜下观察大鼠脑缺血再灌注区的病理形态改变。结果:缺血再灌注组与假手术组相比,血浆IL-1β、TNF-α含量明显增加(P<0.05),E-选择素治疗组血浆IL-1β、TNF-α水平均降低(P<0.05)。光镜和电镜下观察见缺血再灌注组额顶叶皮质和基底节区神经细胞呈缺血性改变,应用人工合成E-选择素后上述区域缺血性改变可明显减轻。结论:应用人工合成E-选择素能减轻大鼠脑缺血再灌注损伤后炎症细胞因子IL-1β和TNF-α的表达,减轻神经细胞缺血性改变,对大鼠脑缺血再灌注损伤具有保护作用。Aim: To investigate the mechanism of the protect role induced by preventive application of artificial synthesized E-selectin in cerebral ischemia-reperfusion injury of rat. Methods: 90 male SD rats weighed 280-320 g were randomly divided into the three groups: (1) sham-operation group, (2) ischemiareperfusion group, (3)artificial synthesized E-selectin treated group, for each group 30 rats. The focal cerebral ischemia-reperfusion models of middle cerebral artery occlusion(MCAO) were made by referring Longa's method. 5 rain before building the models in artificial synthesized E-selectin treated group, E-selectin(10 mg·kg^-1) were injected into femoral vein. At different time(2, 6, 12, 24, 48 and 72 h after reperfusion), phlebotomizing from the heart, mensurating the content of interleukin-1β (IL-1β ),tumor necrosis factor-α (TNF-α) of plasma by the method of ELISA. Then the rats were killed and their brains were excluded, sections were made, and the pathomorphological changes of the neurons in the ischemia-reperfusion area were observed by light microscope and electron microscope. Results: The content of plasma IL-1β,TN-α in ischemia-reperfusion group was higher than those of sham-operation group(P〈0.05). The plasma IL-1β, TNF-α content in treated group was reduced(P〈0.05). The obvious ischemia changes of the cells were shown in the areas of frontal-parietal cortex and basal ganglion in the ischemia-reperfusion group and in the treatment group, the cell injury was obvious reduced by the microscope. Condusions.. The preventive application of artificial synthesized E-selectin obviously reduced the expression of plasma IL-1β, TNF-α in the cerebral ischemia-reperfusion injury of rat, and reduced the ischemia changes of cerebral cell. The artificial synthesized E-selectin has the protective role to the cerebral ischemia-reperfusion injury in treated group.
