摘  要:目的:研究尿多酸肽(CDA-2)对EAC人子宫内膜癌细胞增殖、分化及细胞自分泌的影响。方法:应用MTT法、集落形成实验检测CDA-2对EAC细胞增殖的影响;通过光镜观察和免疫组化鉴定细胞分化;放射免疫法检测细胞自分泌雌二醇(E2)、孕酮(P)。结果:CDA-2使EAC细胞增殖与集落形成均明显受到抑制,并呈剂量依赖性。CDA-2处理72h后:EAC出现细胞分化的形态学改变,免疫组化显示ER、PR表达阴性。EAC细胞无自分泌E2、P。结论:CDA-2不仅能抑制子宫内膜癌EAC细胞增殖,而且能诱导EAC细胞分化。Objective: To study the effect of Uroacitide (CDA-2) on the prnliferation, differentiation and autoerine of human eudometrial carcinoma cell line EAC. Methods: MTT assay and colony formation assay were employed to detect the effect of CDA-2 on cellular survival and proliferation. The differentiation of EAC cell line was ideutified by HE staining and immunoeytochemistry. E2 and P in eudometrial carcinoma cell line EAC were quantified by radioimmunoassay. Results: The cellular survival and proliferation of EAC cell line were remarkably inhibited by CDA-2 and the effect was dose-dependent. After being treated with CDA-2 for 72 hours, HE staining of EAC cells revealed differentiation induced by CDA-2. No expression of ER and PR were observed by immunocylochemistry. No E2 and P were detected in EAC cells. Conclusion: CDA-2 could inhibit proliferation and induce differentiation in human eudometrial carcinoma cell line EAC.
