摘 要:目的:α-亚麻酸具有抗自由基损伤作用,维生素A,C,E也具有抗氧化和清除自由基能力。实验拟进一步验证。亚麻酸分别与维生素E、维生素C、维生素A和维生素A,C,E联合用药对小鼠抗氧化能力和延长果蝇寿命的作用。方法:实验于200610/2007—05在山西医科大学第三医院检验中心(BSL-2)完成。①实验材料:SPF级18~22月龄老年昆明小鼠60只,1.5~2.0月龄青年昆明小鼠10只,雌雄各半:浓度80% α-亚麻酸为河南利诺生化有限公司产品;美国野生型黑翅果蝇400只由山西大学生命科学院提供。②实验分组:抗氧化能力实验分为老龄小鼠。亚麻酸组、α-亚麻酸+维生素E组、α-亚麻酸+维生素C组、α-亚麻酸+维生素A组、α-亚麻酸+维生素ACE组,每组10只,青年小鼠组10。果蝇寿命实验分为。亚麻酸+维生素E组,α-亚麻酸+维生素C组,α-亚麻酸+维生素ACE组及对照组。③实验评估:测小鼠血清中超氧化物歧化酶、谷胱甘肽过氧化物酶及丙二醛,评价其抗氧化能力;各组果蝇平均寿命、最高寿命。结果:70只小鼠伞部进入结果分析。①5个老年用药组的血超氧化物歧化酶含量、谷胱甘肽过氧化物酶含量高于老年对照组(P〈0.05):α-亚麻酸+维生素ACE联合应用组卜述2项指标高于其他4个用药组(P〈0.05)。②5个老年用药组的丙二醛含量低于老年对照组(P〈0.01),与青年对照组差异无显著性(P〉0.05)。③α-亚麻酸与维生素A,C,E联合应用,雌雄性果蝇平均寿命延长率及最高寿命延长率均高于。亚麻酸+维生素C、α-亚麻酸+维生素E组(P〈0.05)。结论:α-亚麻酸与维生素ACE联合应用比分别与。亚麻酸合用能更显著提高小鼠超氧化物歧化酶和谷胱甘肽过氧化物酶、降低丙二醛水平,果蝇寿命更长。AIM: α-linoleic acid can inhibit the damage induced by free radical. Vitamin A, C, and E also possess the effect of antioxidation and eliminating free radical. In this study, we explored the effect of α-linoleic acid plus vitamin E, A, C respectively and ACE on eliminating free radical and prolonging life span of drosophila. METHODS: The experiment was performed at Test Center of the Third Hospital of Shanxi Medical University between October 2006 and May 2007. ①Sixty SPF Kunming mice of 18-22 months old and 10 Kunming mice of 1.5-2 month old with an equal amount of males and females were selected, α - linoleic acid (80%) was product of Henan Linuo Biochem Co., Ltd. 400 wild drosophila melanogaster were provided by College of Life Science, Shanxi University. ②In antioxidation test, 18 22 months old mice were divided into α-linoleic acid group, α-linoleic acid plus vitamin A, C, E respectively and α-linoleic acid plus ACE groups (n=10). In addition, 10 young mice served as young control group. In life test, drosophila melanogaster were divided into α-linoleic acid, α-linoleic acid plus vitamin C, E respectively and ACE groups, and control group. ③Antioxidation ability was estimated by measuring superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and malonaldehyde (MDA). Average life and the maximum life of drosophila were explored. RESULTS: Seventy mice were all included in final analysis. ①Higher SOD and GSH-Px were found in all 5 groups treated with α -linoleic acid compared with old control mice (P 〈 0.05), especially in α-linoleic acid plus ACE mice (P 〈 0.05). ②MDA con- centration in all five groups treated with α- linoleic acid was significantly lower than old control mice (P 〈 0.05), but it was not significantly different compared with young control mice (P 〉 0.05). ③α-linoleic acid plus E, C, A groups significantly prolonged the average life and the maximum life of drosophila than α-linoleic acid plus C, and α -linole
