摘 要:目的研究1,25-二羟维生素D3对豚鼠实验性变态反应性脑脊髓炎(EAE)外周血淋巴细胞亚群的影响,探讨1,25-二羟维生素D3防治EAE的免疫调节机制。方法采用粗制碱性髓鞘蛋白(coarsemyelin basic protein,CMBP)加福氏完全佐剂皮下注射的方法建立EAE模型。将50只豚鼠随机分成5组:正常对照组、EAE对照组、EAE低剂量治疗组、EAE中剂量治疗组和EAE高剂量治疗组,每组10只,造模前3d正常对照组和EAE对照组每天灌喂0.4ml花生油,EAE低、中、高剂量治疗组分别在造模前3d开始灌喂含1,25-二羟维生素D30.5μg·kg-1.d-1,1.25μg·kg-1.d-1,2.5μg·kg-1.d-1的花生油0.4ml,至实验结束。流式细胞仪法测定高峰期外周血T细胞亚群。结果EAE对照组外周血CD4+、CD8+含量及CD4+/CD8+比值较正常对照组差异有统计学意义(P<0.05);EAE低、中、高剂量治疗组外周血CD4+、CD8+含量及CD4+/CD8+比值较EAE对照组差异有统计学意义(P<0.05)。结论(1)EAE豚鼠外周血CD4+、CD8+含量下降、CD4+/CD8+比值升高,存在T细胞亚群失衡;(2)1,25-二羟维生素D3能提高外周血CD4+和CD8+含量、降低CD4+/CD8+比值,具有调节T细胞亚群失衡和纠正CD4+/CD8+比值倒置的作用。Objective To investigate the influence of 1, 25 - dihydrovitamin D3 on T lymphocyte subsets of peripheral blood in guinea pigs with experimental autoimmune encephalomyelitis (EAE) so as to explore into its immune regulating mechanisms in prevention and treatment of EAE. Methods The Guinea pig models with EAE were set up by subcutaneous injection of coarse myelin basic protein (CMBP) with Freund's Complete Adjuvant. Totally. 50 Guinea pigs were divided randomly into five groups; the nomral, EAE control, 1, 25 - dihydrovitamin D3 low dose, medium dose and high dose, with 10 in each group. Three days before the model establishment, the animals in the normal group and EAE control group were by gavage given peanut oil 0. 4 ml daily, and the animals in the 1, 25 - dihydrovitamin D3 low dose, medium dose and high dose groups were by gavage given the peanut oil 0.4 ml with1, 25 - dihydrovitamin D3 0.5μg·kg^-1·d^-1,1.25 μg·kg^-1·d^-1, 2. 5μg·kg^-1·d^-1 respectively, until the end of the experiment. The peak levels of T lymphocyte subsets of peripheral blood of the animals in all five groups were detected with Flow Cytometer. Results There were significant differences in percentages of CD4^+ and CD8^+ T lymphocytes, and the ratio of CD4^+ / CD8^+ of peripheral blood between the EAE control group and the normal group ( P 〈 0. 05 ). And the percentages of CD4^+ and CD8^+ T lymphocytes, and the ratio of CD4^+ / CD^+ of peripheral blood in EAE control group were deferent from those of 1, 25 - dihydrovitamin D3 low dose, medium dose and high dose groups ( P 〈 0. 05 ). Conclusion ( 1 ) The levels of CD4^+ and CD8^+ T lymphocytes in peripheral blood of guinea pig with EAE decrease, and the ratio of CD4^+/CD8^+ increases, with an imbalance of T lymphocyte subsets; (2) 1, 25 -dihydrovitamin D3 could increase the levels of CD4^+ and CD8^+ , decrease the ratio of CD4^+ / CD8^+ , with a role of regulating the imbalance of T lymphocyte subsets and correcting the inv
