摘  要:目的:观察糖皮质激素受体阻断剂(RU486)对EAE大鼠的影响。方法:雌性Wistar大鼠23只分成三组:对照(Control)组、模型(EAE)组、RU486组,采用豚鼠脊髓匀浆/完全弗氏佐剂(GPSCH/CFA)免疫Wistar大鼠建立EAE模型,采用行为学变化观察动物的发病情况,常规HE和Kluver&Barrera染色法观察中枢神经系统(CNS)的病理变化情况。结果:①EAE组7/8大鼠出现典型的EAE行为学改变、CNS炎性细胞浸润(Okuda评分)和髓鞘脱失。②RU486组3/9大鼠出现行为学改变、CNS炎性细胞浸润和髓鞘脱失,均重于模型组(P<0.01)。结论:RU486未增加大鼠EAE的发病率,但可使EAE大鼠的行为学改变、CNS炎性细胞浸润和髓鞘脱失加重。Objective: To observe the effect of glueocortieoid receptor antagon (ru486) on experimental allergic encephalomyelitis in Wistar rats.Methods: Twenty three female Wistar rats were divided into three groups: control group, model group( EAE group) and RU486 group.EAE was induced by subcutaneous injection into the four footpats with guinea pig spinal cord homogenate(GPSCH) emulsified in complete Freund' s adjuvant(CFA). The diet, movement and body weight of Wistar rats were daily examined, CNS tissues were stained with hematoxylineosin and Kluver&Barrera. Results: (1)Seven of eight rats in EAE group demonstrated neurological signs of typical EAE: parenchymal infiltration, perivascular culling with mononuelear cells, and demyelination in the white matter. (2)Three of nine in RU486 group demonstrated neurelogical signs of typical EAE, with parenchymal infiltration, perivascular culling with mononuclear cells, and demyelination in the white matter. All the phenomena were more serious than in EAE group( P 〈 0.01 ). Conclusion: RU486 could not increase the incidence of EAE, but it makes neurological signs,inflammation and demyelination more serious.
