细胞凋亡过程中形态学变化

摘　　要：用倒置显微镜、光镜及透射电镜观察了人食管癌Ｅｃａ１０９细胞经顺铂作用后的形态学变化及细胞死亡特征。结果显示：肿瘤细胞皱缩出芽、核裂解及凋亡小体形成。电镜下凋亡细胞表面的微绒毛完全消失，代之以圆形、半圆形突起。一些细胞伸出多个不规则伪足样的突起。在消化收集药物作用后的细胞时，发现仍贴壁的细胞回缩比未用药对照细胞回缩的速度缓慢。结果提示：在细胞凋亡及其形态变化的过程中，可能伴随着细胞胞质的异常运动。Morphological changes of cisplatin(DDP) induced apoptosis in human esophageal carcinoma Eca 109 cells were observed with inverted,light and electron microscopes.The results showed that the retraction,budding,nuclear fragmentation and the formation of apoptotic bodies were apparent.Under the electron microscope,microvilli on the surface of apoptotic cells disappeared and was substituted by circular or semicircular protuberances.In addition.irregular microspikes or pseudopodia stretched from the surface of some apoptotic cells.When the cells treated with trypsin EDTA in PBS were collected,it was found that these cells which were still adherent to the culture plates after they were treated with DDP retracted,and the retraction was slower than that of the control cells.The results suggest that the process and the morphological changes of apoptosis may be accompanied with the disrupture of cytoskelet and the abnormal cytoplasmic movement.
