摘 要:优化了提取Rhizobium sp.N613胞外多糖(REPS)的工艺条件,以发酵液pH值、乙醇浓度以及醇沉时间为因子,采用响应面法试验获得了REPS的最佳提取条件,即pH5.9,乙醇浓度74%,醇沉时间16.5h。在该条件下,胞外多糖提取率为9.28±0.06g/L,其纯度达97%,收率达93.6%。此外,还进行了REPS的特性粘度、急性毒性测试和蓄积毒性实验,结果表明该多糖具有粘度高、无毒性的特点,在食品医药领域有着广泛的应用前景。The process of extracting exopolysaccharide (REPS) from Rhizobium sp. N613 was optimized. The optimized conditions of extracting REPS were obtained by response surface methodology. The pH value is 5.9, concentration of ethanols is 74% and the precipitation time is 16. 5 h, respectively. Under those conditions, the extraction yield was 9.28 ±0.06g/L, the purity was up to 97% and the extraction rate was 93. 6%. At the same time, the thesis carried out the test of the viscosity, acute toxicity and accumulative toxicity of REPS. It is showed that REPS has high viscosity and nontoxicity. So it is more valuable for the application for food and medicine.
