摘 要:目的:观察8Hz90dB次声作用对大鼠海马PKA表达及CREB磷酸化水平的影响。方法:采用免疫组织化学方法,观察8Hz90dB次声作用不同时间点大鼠海马PKA表达及p-CREB-1(Ser-133)磷酸化水平。结果:8Hz90dB次声作用后,大鼠海马CA1区各时间点PKA表达均上调,相应时间点CREB磷酸化水平亦上调,两者之间的变化呈显著正相关。CA3区除7d组PKA表达上调、CREB磷酸化水平亦上调外,两者之间总变化趋势为显著负相关。DG各时间点PKA表达与CREB磷酸化水平变化之间无显著相关性。结论:8Hz90dB次声作用对大鼠海马各区PKA表达及CREB磷酸化水平均有不同程度的影响,提示次声对学习记忆功能影响的机制可能涉及对cAMP浓度增高→PKA激活→CREB磷酸化→CRE依赖性转录这一分子链产生了干扰作用。Objective:To observe effects of infrasound on PKA expression and CREB phosphorylation in hippocampus of rats.Method:PKA expression and CREB phosphorylation in hippocampal areas were studied with immunohistochemistry techniques at different time points after the rats were exposed to infrasound of 8Hz 90dB.Result:When the rats were exposed to 8Hz 90dB infrasound, PKA expression and CREB phosphorylation were positively correlated in CA1, but negatively correlated in CA3 and not correlated in DG at all the time points. Obviously PKA expression and p-CREB-1(Ser-133) phosphorylation in different regions had different responses to 8Hz 90dB infrasound.Conclusion:In the experiment, the relationship between PKA expression and p-CREB-l(Ser-133) phosphorylation wasn't parallel in some areas or at some time points. The effects of 8Hz 90dB infrasound on learning and memory function of hippocampus may be due to the mechanism of disturbing the molecule chains, increasing cAMP concentration -activating PKA-CREB phosphorylation- CRE dependent transcription.
